尾加压素Ⅱ促进毛乳头细胞增殖及分泌血管内皮生长因子的实验研究

摘　　要：目的初步研究尾加压素Ⅱ(urotensinⅡ,UⅡ)对体外培养的毛乳头细胞增殖以及分泌血管内皮生长因子(VEGF)的影响.方法采用一步酶法分离原代毛乳头细胞并进行体外培养,取2~4代细胞用于实验.用含不同浓度UⅡ的DMEM培养基培养毛乳头细胞,分别在0、24、48、72 h采用CCK-8法检测毛乳头细胞的增殖情况,在72 h采用酶联免疫吸附试验(ELISA)检测毛乳头细胞培养液上清中VEGF蛋白含量.结果与对照组相比,10^(-6)、10^(-7)、10^(-8) mol/L UⅡ可促进毛乳头细胞增殖,10^(-6)、10^(-7)mol/L UⅡ亦可明显干预细胞VEGF的表达.结论UⅡ可促进毛乳头细胞增殖及分泌VEGF.Objective To investigate the effect of urotensin Ⅱ(U Ⅱ) on dermal papilla cells’ proliferation and secretion of vascular endothelial growth factor(VEGF).Methods The primary dermal papilla cells were isolated from rat hair follicles by one-step enzymatic method and cultured in vitro,then the 2-4 generation cells were selected for the experiment.The dermal papilla cells were cultured in DMEM medium containing different concentrations of UⅡ,and CCK-8 method was adopted to detect the proliferation of the cells at 0,24,48 and 72 h,and the level of VEGF in the supernatant was detected by ELISA at 72 h.Results Compared with the control group,10-6,10-7 and 10-8 mol/L UⅡ could promote the proliferation of dermal papilla cells,and 10-6,10-7 mol/L UⅡ could also significantly influence the expression of VEGF protein of dermal papilla cell.Conclusion Urotensin Ⅱ may promote the proliferation and VEGF protein secretion of dermal papilla cell.
