摘 要:目的探讨西洋参总皂苷对急性血瘀模型大鼠的作用,为其临床应用提供科学依据。方法选取健康大鼠60只,随机分为正常对照组、模型组、阳性对照组及西洋参总皂苷低、中、高剂量(100、200、400 mg·kg^(-1))组,连续给药8 d,采用皮下注射肾上腺素复加冰水浴的方法,复制急性血瘀模型大鼠,观察西洋参总皂苷对急性血瘀模型大鼠血液流变学、凝血四项及血浆中α颗粒膜蛋白(GMP-140)、血管性血友病因子(vWF)含量的影响。结果与模型组相比,西洋参总皂苷使急性血瘀模型大鼠的全血低、中、高切黏度及血浆黏度显著降低;使凝血酶原时间、活化部分凝血酶原时间、凝血酶时间显著升高,纤维蛋白原显著降低;使GMP-140、vWF含量显著减少。结论西洋参总皂苷通过调节血液流变学指标和凝血系统功能,降低血液黏度、抑制血小板活化和黏附聚集,明显改善急性血瘀症状,这可能与其降低血浆中GMP-140、vWF的含量有关。Objective To determine the effect of total saponins of American ginseng(AG)on acute blood stasis rat model,and to provide scientific basis for clincal application.Methods Totally 60 healthy rats were randomly divided into a normal control group,a model group,a positive control group and the AG total saponins low-dose,medium-dose and high-dose(100,200,and 400 mg·kg^(-1))groups with continuous preventive administration for 8 days.The model of acute blood stasis in the rats were replicated by the subcutaneous injection of epinephrine with ice water bath.The effects of total saponins of AG on the hemorheology,coagulation,platelet granular membrane protein 140(GMP-140)and von willebrand factor(vWF)in the plasma of rats with acute blood stasis were observed.Results Compared with the model group,total saponins of AG significantly decreased the whole blood viscosity and plasma viscosity,while increased the prothrombin time,the activated partial prothrombin time and thrombin time.The content of GMP-140 and vWF was significantly decreased.Conclusion The total saponins of AG can significantly improve the symptoms of acute blood stasis by regulating the hemorheological indexes and function of the coagulation system,reducing blood viscosity,inhibiting platelet activation and adhesion aggregation,which may be related to the down regulation of the content of GMP-140 and vWF in the plasma.
