通窍活血汤对大脑中动脉栓塞模型大鼠神经功能及HPA轴功能影响

摘　　要：目的:探讨研究通窍活血汤对大脑中动脉栓塞模型大鼠神经功能及下丘脑-垂体-肾上腺皮质轴(Hypothalamus-pituitary-adrenal cortex,HPA)功能的影响。方法:选取无特定病原体级SD雄性大鼠48只为研究对象,随机分为通窍活血汤高、中、低剂量组、对照组、假手术组和模型组,每组8只,造模成功后,高、中、低剂量组按照12.04、6.02和3.01g/kg标准分别灌胃通窍活血汤,其余组灌胃等量生理盐水,检测大鼠的大便、精神、尾色等,在手术后3h、6h、1d、6d和7d对大鼠神经功能进行测评,使用放免法检测大鼠血清促肾上腺皮质激素(Adreno-cortico-tropic-hormone,ACTH)、皮质酮(Corticosterone,CORT)和血浆促肾上腺皮质激素释放激素(Corticotropin releasing hormone,CRH)水平。结果:模型组大鼠手术后3h、6h、1d、6d和7d神经功能评分均高于对照组,差异有统计学意义(P<0.05),通窍活血汤高、中剂量组大鼠手术后6d和7d神经功能评分均低于模型组,差异有统计学意义(P<0.05),通窍活血汤低剂量组大鼠手术后7d神经功能评分低于模型组,差异有统计学意义(P<0.05);模型组大鼠血清ACTH和血浆CRH水平均低于对照组,血清CORT水平高于对照组,差异有统计学意义(P<0.05),通窍活血汤中、高剂量组血清ACTH和血浆CRH水平均高于模型组,血清CORT水平低于模型组,差异有统计学意义(P<0.05),高剂量组血清ACTH和血浆CRH水平均高于低剂量组,血清CORT水平低于低剂量组,差异有统计学意义(P<0.05),假手术组与对照组大鼠血清CORT、ACTH和血浆CRH水平差异无统计学意义(P>0.05)。结论:通窍活血汤可改善大脑中动脉栓塞模型大鼠神经功能,调节HPA轴功能。Objective:To investigate the effect of Tongqiao Huoxue Decoction on the neurological function and the function of hypothalamic-pituitary-adrenal cortex(HPA)for rats with middle cerebral artery occlusion.Methods:Forty-eight SD male rats without specific pathogens were randomly divided into high-dose,medium-dose,and low-dose groups of Tongqiao Huoxue Decoction,controlled group,sham operation group and model group,with 8 rats in each group.The high,middle and low dose groups were given Tongfu Huoxue Decoction according to the standards of 12.04,6.02 and 3.01 g/kg.The other groups were given the same amount of normal saline to detect the stool,spirit and tail color of the rats,3 h and 6 h after operation.1 d,6 d and 7 d were used to measure the neurological function of rats,and the serum adrenocortin-tropic-hormone(ACTH),corticosterone(CORT)and plasma adrenocorticotropic hormone were detected by radioimmunoassay.The level of corticotropin releasing hormone(CRH)was released.Results:The neurological scores of the model group were higher than those of the controlled group at 3 h,6 h,1 d,6 d and 7 d after operation(P<0.05).The rats in Tongfu Huoxue Decoction high and middle dose group were 6 d and 7 d after operation.The neurological function scores were lower than the model group,and the difference was statistically significant(P<0.05).The neurological function score of the low-dose group of Tongqiao Huoxue Decoction group was lower than that of the model group(P<0.05).The levels of serum ACTH and plasma CRH in the model group were lower than those in the controlled group,and the serum CORT level was higher than that in the controlled group.The difference was statistically significant(P<0.05).The serum ACTH and plasma CRH levels in the middle and high dose groups of Tongqiao Huoxue Decoction.Above the model group,the serum CORT level was lower than the model group,the difference was statistically significant(P<0.05).The serum ACTH and plasma CRH levels in the high dose group were higher than the low dose group,and the s
