摘  要:目的:通过比较不同肾虚模型和补肾中药治疗后各组大鼠唾液腺AQP1、AQP2、AQP5的变化,探索肾与唾相关的内在分子机制及补肾药的干预作用。方法:将清洁级SD大鼠40只按照随机数字表法分成正常对照组、肾阴虚模型组、肾阳虚模型组、六味地黄丸组、金匮肾气丸组,每组8只。分别采用灌胃甲状腺片(45mg/100g)混悬液和肌肉注射氢化可的松(2.5mg/100g)方法制备肾阴虚和肾阳虚模型组,并分组进行药物干预治疗。比较各组大鼠的一般状况、唾液流率、唾液渗透压、颌下腺病理组织、颌下腺AQP1、AQP2、AQP5蛋白表达变化情况。结果:与正常对照组比较,肾阴虚和肾阳虚证动物模型大鼠体重减轻,唾液流率降低,渗透压有升高的趋势,颌下腺组织中AQP1、AQP2、AQP5蛋白表达减少。应用补肾药进行干预治疗后,G干预组相应指标的变化得到了改善。结论:不同肾虚状态下唾液分泌减少,其发生的内在机制可能与唾液腺的水通道蛋白表达降低有关,补肾药可以改善肾虚状态下唾液分泌异常的状态。Objective:By comparing the changes of salivary glands AQP1,AQP2,and AQP5 after different kidney deficiency models and kidney-tonifying Chinese medicine treatment,the internal molecular mechanism of kidney-saliva correlation and the intervention effect of kidney-tonifying drugs were explored.Methods:Forty SD rats with clean grade were divided into normal controlled group with kidney-Yin deficiency model group with kidney-Yang deficiency model group with Liuwei Dihuang Pill group with Jinkui Shenqi Pill group with 8 rats in each group according to random number table.The model groups of kidney-Yang deficiency and kidney-Yang deficiency were prepared by gavage Thyroid Tablet(45 mg/100 g)suspension and intramuscular hydrocortisone(2.5 mg/100 g)respectively,and were divided into groups for drug intervention.The general condition,salivary flow rate,salivary osmotic pressure,and the expression of AQP1,AQP2 and AQP5 proteins in the submandibular gland were compared in each group.Results:The internal mechanism of decreased salivary secretion in different deficient states of the kidney may be related to decreased expression of aquaporin in salivary glands,and tonifying kidney drugs can improve abnormal salivary secretion in deficient states of the kidney.
