摘  要:旨在对口蹄疫A型病毒(FMDV)3种毒株制备的不同抗原进行反应性比较分析,筛选最合适的免疫学诊断抗原,为进一步研究利用该抗原进行血清学诊断奠定基础。分别选择AF/72毒株、Re-A/WH/09毒株制备纯化146S抗原和VP1表达蛋白以及A/GD/MM/13毒株VP1表达蛋白作为备选抗原,利用间接ELISA检测方法与胶体金免疫层析检测方法(GICA),对100份背景清晰的田间样本血清进行平行检测,比较不同抗原的反应性。结果显示,A/GD/MM/13株抗原活性最低,间接ELISA方法检测AF/72株、Re-A/WH/09株纯化146S抗原和VP1表达蛋白对于A型阳性血清敏感性分别为90.00%、86.67%、78.33%和76.67%,与O型、Asia1型阳性血清和阴性血清特异性结果分别为95.00%、87.50%、75.00%和70.00%。GICA方法检测AF/72株、Re-A/WH/09株纯化146S抗原对于A型阳性血清敏感性分别为95.00%和83.33%,与O型、Asia1型阳性血清和阴性血清特异性结果分别为92.50%和87.50%。本研究通过FMDV-A型不同毒株抗原反应性的比较试验证实,AF/72株纯化146S抗原在间接ELISA和GICA 2种方法中均显示更好的敏感性、特异性,且在纯化过程中,收获量更多、浓度更高,为最合适的诊断用抗原,所建立的诊断方法可获得满意的检测结果。To compare and analyze the reactivity of different antigens prepared by three strains of foot-and-mouth disease virus type A(FMDV),and to choose the most suitable diagnostic antigens, so as to lay a foundation for the study of serological diagnosis.The purified 146 S protein of AF/72,VP1 expression protein of Re-A/WH/09,and VP1 expression protein of A/GD/MM/13 were selected as the candidate antigens.Indirect ELISA and GICA methods were established, and 100 serum samples with clear background were tested.The results showed that the antigen activity of A/GD/MM/13 strain was the lowest.Indirect ELISA revealed that the sensitivity of 146 S and VP1 expression protein purified from AF/72 and Re-A/WH/09 to FMDV type A positive serum was 90.00%,86.67%,78.33% and 76.67%,respectively.The specificity of them to the positive serum and the negative serum of type O and type Asia1 was 95.00%,87.50%,75.00% and 70.00%,respectively.In GICA,the sensitivity of 146 Spurified by AF/72 and Re-A/WH/09 was95.00%and 83.33%,respectively,and the specificity was 92.50%and 87.50%,respectively.In this study,the comparison of antigen reactivity of different strains of FMDV type A confirmed that146 Spurified by AF/72 showed better sensitivity and specificity in both indirect ELISA and GICA,and in the process of purification,the concentration of 146 Spurified by AF/72 is higher.As the most appropriate antigen,the diagnosis method established by using 146 Spurified by AF/72 can obtain a satisfactory result.
