摘 要:目的通过分析大鼠丙泊酚麻醉前后工作记忆编码阶段海马和内侧前额叶皮层局部场电位(LFPs)的能量密度和神经回路频谱相干性,研究丙泊酚麻醉对工作记忆编码的影响。方法以8只SD大鼠为研究对象,在大鼠内侧前额叶皮层和腹侧海马各植入16通道微电极阵列。利用植入微电极阵列采集LFPs记录SD大鼠麻醉前后Y迷宫工作记忆行为学结果。采用短时傅里叶变换(STFT)对LFPs进行时频分析,应用时变频谱相干方法计算相干系数。结果大鼠在麻醉后12 h工作记忆行为学正确率显著低于麻醉前(P<0.01),麻醉后24 h恢复到正常水平。丙泊酚麻醉后工作记忆损伤大鼠内侧前额叶皮层theta频段平均能量密度显著低于麻醉前(P<0.05);麻醉后gamma频段平均能量密度显著低于麻醉前(P<0.01),腹侧海马theta频段平均能量密度低于麻醉前(P<0.05);gamma频段平均能量密度在麻醉后显著性低于麻醉前(P<0.01)。丙泊酚麻醉后海马-前额叶皮层theta相干系数显著低于麻醉前(P<0.001),gamma相干系数在麻醉前和麻醉后有显著性差异(P<0.01)。结论丙泊酚损伤工作记忆是暂时性的,且这种暂时性的损伤可能是抑制工作记忆编码阶段海马和前额叶皮层频率同步造成。Objective To study the effect of propofol anesthesia on working memory encoding from the perspective of theta and gamma frequency energy density and coherence of local field potentials(LFPs)in hippocampus and prefrontal cortex,the two key regions of working memory encoding.Methods Eight SD rats were used as the test objects and the microelectrode arrays were implanted in medial prefrontal cortex(mPFC)and ventral hippocampus(vHPC)of rats.By using the technique of microelectrode array recording of awake animals in vivo,the LFPs were recorded while rats performing working memory tasks in Y-maze before and after anesthesia.Then the frequency power spectrum of LFPs was calculated by short time Fourier transform(STFT)and coherence between vHPC and mPFC.The statistical comparison of power and coherence between the control and the propofol group were calculated.Results The performance accuracy 12 h after propofol anesthesia declined as compared with that before anesthesia(P<0.01),but recovered 24 h after propofol.The power of theta band in mPFC 12 h after propofol anesthesia was significantly lower than that before anesthesia(P<0.05);while those of vHPC(P<0.01).The reduction of power 12 h after propofol anesthesia could be found in gamma band both in vHPC and mPFC.vHPC-mPFC theta coherence(P<0.01)before and 12 h after anesthesia;and gamma coherence 12 h after anesthesiawas significantly lower than that before anesthesia(P<0.01).Conclusion Propofol anesthesia reversibly impaired the working memory in adult rats.The impairment may result from the decreased oscillations synchrony between vHPC and mPFC after propofol anesthesia.
