摘 要:目的:观察瓜子金皂苷己(Polygalasaponin F,PGSF)是否影响氧糖剥夺/复氧(Oxygen and glucose deprivation/reoxygenation,OGD/R)处理的BV-2小胶质细胞炎性细胞因子及细胞因子信号抑制因子3(Suppressors of cytokine signaling3,SOCS3)表达,初步探讨PGSF对抗OGD/R炎症反应的机制。方法:构建BV-2小胶质细胞OGD/R体外模型,分为正常对照组、模型组、PGSF低剂量组、PGSF中剂量组及PGSF高剂量组。BV-2细胞氧糖剥夺2 h、复糖复氧6 h后提取总RNA,通过实时荧光定量多聚酶链式反应检测BV-2细胞炎性因子TNF-α、IL-1β、IL-6以及SOCS3的基因表达。结果:I/R后BV-2细胞中炎性因子TNF-α、IL-1β、IL-6及SOCS3的mRNA表达明显增加;给予PGSF处理可以逆转上述观察指标的改变。结论:PGSF在体外实验中具有抑制神经炎症的作用,SOCS3信号通路可能参与介导了PGSF的抗炎作用。Objective:To investigate the effects of polygalasaponin F(PGSF)on the expression of inflammatory cytokines and suppressors of cytokine signaling 3(SOCS3)in BV-2 microglia treated with oxygen and glucose deprivation/reoxygenation(OGD/R),and to preliminarily explore the mechanism of PGSF against OGD/R inflammation.Methods:OGD/R models of BV-2 microglia were established in vitro and divided into the normal control group,the model group,the low-dose PGSF group,the medium-dose PGSF group and the high-dose PGSF group.Total RNA was extracted from BV-2 cells after oxygen glucose deprivation for 2 h and reoxygenation for 6 h,with the gene expressions of TNF-α,IL-1β,IL-6 and SOCS3 in BV-2 cells detected by real-time fluorescent quantitative polymerase chain reaction(qPCR).Results:The mRNA expressions of TNF-α,IL-1β,IL-6 and SOCS3 in BV-2 cells were significantly increased after OGD/R insult and PGSF treatment could reverse these changes.Conclusion:PGSF can inhibit neuroinflammation through in vitro experiments,and the SOCS3 signaling pathway may be involved in the anti-inflammatory effect of PGSF.
