摘 要:目的:探究益肾通络方对大鼠勃起功能障碍(ED)及一氧化氮-环磷酸鸟苷(NO-cGMP)通路的影响。方法:构建ED模型大鼠,随机分为模型组,益肾通络方低、中、高剂量组,以及西地那非组,每组12只;另取12只SD大鼠设为对照组。采用阿扑吗啡实验检测大鼠勃起功能,测定最大阴茎海绵体内压(Max ICP)/平均动脉压(MAP),ELISA检测血清睾酮水平、阴茎组织一氧化氮合酶(NOS)活性及NO和cGMP水平,Western blot法检测阴茎组织NOS和鸟苷酸环化酶(GC)的表达。结果:与对照组比较,模型组大鼠勃起次数显著减少,Max ICP/MAP、血清睾酮水平、阴茎组织NOS活性、NO和cGMP水平及NOS和GC蛋白表达均显著降低(P<0.05),勃起潜伏期显著延长(P<0.05);与模型组比较,益肾通络方低、中、高剂量组和西地那非组大鼠勃起次数显著增多,Max ICP/MAP、血清睾酮水平、阴茎组织NOS活性、NO和cGMP水平及NOS和GC蛋白表达均显著升高(P<0.05),勃起潜伏期显著缩短(P<0.05),且益肾通络方各组之间呈剂量依赖性(P<0.05);益肾通络方高剂量组与西地那非组相比,各指标的差异无统计学显著性(P>0.05)。结论:益肾通络方可提高血清睾酮水平并减轻大鼠ED,这可能是通过上调NO-cGMP通路实现的。AIM:To explore the effect of Yishentongluo decoction(YSTL)on erectile dysfunction(ED)and nitric oxide-cyclic guanosine monophosphate(NO-cGMP)pathway in rats.METHODS:An ED rat model was estab-lished,and the rats were randomly divided into model group,low-,medium-and high-dose YSTL groups and sildenafil group,with 12 rats in each group.Another 12 SD rats were selected as control group.After treatment,apomorphine test was used to detect erectile function,and the maximum intracavernous pressure(Max ICP)/mean arterial pressure(MAP)was measured.The serum level of testosterone,and nitric oxide synthase(NOS)activity and NO/cGMP levels in penile tissue were measured by ELISA.The expression of NOS and guanosine cyclase(GC)in penile tissue of rats was deter-mined by Western blot.RESULTS:Compared with control group,the number of erections,Max ICP/MAP,the serum level of testosterone,the activity of NOS in penile tissue,the levels of NO and cGMP in penile tissue,and the expression of NOS and GC in penile tissue of model group were significantly decreased(P<0.05),while the latent period of erection was significantly increased(P<0.05).Compared with model group,the number of erections,Max ICP/MAP,the serum level of testosterone,the activity of NOS in penile tissue,the levels of NO and cGMP in penile tissue,and the expression of NOS and GC in penile tissue of low-,medium-and high-dose YSTL groups and sildenafil group were significantly in-creased(P<0.05),while the latent period of erection was significantly decreased(P<0.05).A dose-dependent relation-ship among YSTL groups was observed(P<0.05).No significant difference in high-dose YSTL group and sildenafil group was found(P>0.05).CONCLUSION:Yishentongluo decoction elevates the serum level of testosterone and attenuates ED in rats,which may be achieved by up-regulating the NO-cGMP pathway.
