摘 要:为探究蒙古黄芪(Astragalus mongholicus Bunge)种子在控制劣变(Controlled deterioration treatment,CDT)处理后种子活力及生理生化特性,以含水量为3.7%、6.6%、9.8%的蒙古黄芪种子为材料,采用(53±0.5)℃高温对其进行控制劣变处理,测定劣变后蒙古黄芪种子发芽指标及生理生化指标。结果表明:随劣变时间延长,种子发芽率和发芽指数下降、超氧阴离子和过氧化氢含量升高、超氧化物歧化酶(SOD)活性上升,抗坏血酸过氧化物酶(APX)、脱氢抗坏血酸还原酶(DHAR)、单脱氢抗坏血酸还原酶(MDHAR)、谷胱甘肽还原酶(GR)活性均下降、丙二醛积累增多、可溶性糖和可溶性蛋白含量下降;含水量3.7%的种子上述指标变化幅度较含水量9.8%的种子小,而含水量6.6%的种子居中;低含水量蒙古黄芪种子在劣变过程中抗氧化能力更强,种子活力更高。因此,为保证优质蒙古黄芪种质资源的可持续利用,在种子贮藏时可以把含水量降到3.7%左右甚至更低。The research aims to explore the seed vigor and physiological and biochemical characteristics of Astragalus mongholicus Bunge seeds after controlled deterioration treatment(CDT). Astragalus mongholicus seeds with water content of 3.7%, 6.6%, and 9.8% were used as materials, and the seeds were treated with(53±0.5) ℃ high temperature to control their deterioration, and the germination physiological and biochemical indexes of Mongolian Astragalus seeds after deterioration were determined. The experimental results showed that with the extension of the deterioration time, the seed germination rate and germination index decreased, the content of superoxide anion and hydrogen peroxide increased, the activity of superoxide dismutase(SOD) increased, ascorbate peroxidase(APX), dehydroascorbate reductase(DHAR), monodehydroascorbate reductase(MDHAR), and glutathione reductase(GR) activities decreased, malondialdehyde accumulation increased, soluble sugar and soluble protein content decreased. The change amplitude of above indicators of seeds with 3.7% water content was smaller than that of seeds with 9.8% water content, while the seeds with 6.6% water content were in the middle. Low-water content Mongolian Astragalus seeds had stronger antioxidant capacity and higher seed vigor during deterioration. Therefore, in order to ensure the sustainable utilization of high-quality Mongolian Astragalus germplasm resources, the water content can be reduced to about 3.7% or even lower during seed storage.
