摘 要:目的:联合GEO数据库使用网络药理学探讨四妙勇安汤治疗类风湿关节炎的主要成分和可能的作用机制。方法:通过GEO芯片数据挖掘OMIM数据库和Genecards数据库,检索收集类风湿关节炎靶点;通过TCMSP数据库,以生物利用度>30%,类药性>0.18为筛选条件,收集四妙勇安汤的活性成分及对应靶点。将四妙勇安汤靶点与类风湿关节炎靶点进行匹配获取靶点基因,使用Cytoscape 3.7.2软件构建蛋白-蛋白相互作用网络,并进行拓扑分析筛选核心靶点。使用R软件进行GO分析和KEGG通路分析。结果:四妙勇安汤有效成分共126个(如山柰酚、槲皮素、豆甾醇、β-谷甾醇、谷甾醇等),对应靶点2044个;类风湿关节炎靶点4360个;中药-疾病靶点匹配获得潜在靶点165个。GO分析生物过程获得通路2329个,细胞组分获得通路89个,分子功能获得通路163个;KEGG通路分析共获得通路160个。结论:本研究为更深层次探究四妙勇安汤及所含四妙勇安汤药物成分方剂治疗类风湿关节炎作用机制奠定了基础。Objective:To explore the main components and possible mechanism of Simiao Yong’an Tang(四妙勇安汤)in the treatment of rheumatoid arthritis(RA)using network pharmacologycombined with GEO database.Methods:GEO chip database was used to mine the OMIM database and Genecards database,which were used to collect rheumatoid arthritis targets.Through TCMSP database,the bioactive components and corresponding targets of Simiao Yong’an Tang were collected under the screening conditions of bioavailability being over 30%and generic property over 0.18.The Simiao Yong’an Decoction target was matched with rheumatoid arthritis target to obtain target gene.The PPI network was constructed using software Cytoscape 3.7.2,and the core target was screened by topology analysis.Software R was used to analyze GO biological function and KEGG pathway enrichment.Results:There were 126 effective components in Simiao Yong’an Tang(such as kaempferol,quercetin,stigmasterol,β-sitosterol,sitosterol etc.).There were 2044 corresponding targets,4360 rheumatoid arthritis targets and 165 potential targets obtained by Chinese medicine-disease target matching.Go analysis showed that there were 2329 pathways in biological process,89 pathways in cell component and 163 pathways in molecular function.A total of 160 pathways were obtained by KEGG pathway analysis.Conclusion:This study laid a foundation for further exploring the mechanism of Simiao Yong’an Tang in the treatment of rheumatoid arthritis.
