基于IBDV悬浮培养制备的ND-IB-IBD三联灭活疫苗免疫效力评估

摘  要:为了评估DF-1细胞悬浮培养增殖的IBDV BJQ902抗原液的病毒滴度和基于此抗原制备的鸡新城疫-传染性支气管炎-传染性法氏囊病三联灭活疫苗的物理性状和免疫效力,将IBDV BJQ902株病毒接种悬浮培养的DF-1细胞制备IBDV抗原液,测定抗原液病毒滴度,将此抗原液与常规方法制备的ND La Sota株和IB M41株抗原液,进行适当浓缩、灭活制备ND-IB-IBD三联灭活疫苗测定其物理性状,用ND-IB-IBD三联灭活疫苗以不同剂量免疫试验鸡,测定其免疫效果。结果显示:IBDV BJQ902株病毒接种微载体悬浮的DF-1细胞后,收获的抗原液毒滴度可达107.78.5TCID50/0.1 m L;制备的ND-IB-IBD三联苗性状稳定;以0.1 mL/只的剂量免疫鸡只即可获得良好保护。研究结果为新型ND-IB-IBD三联疫苗的研制提供理论依据。In order to evaluate the viral titer of IBDV BJQ902 antigen proliferated by DF-1 cell suspension culture and the physical properties and immune efficacy of the ND-IB-IBD triple inactivated vaccine, IBDV BJQ902 strain was inoculated to the suspension cultured DF-1 cells to prepare IBDV antigen, and the viral titer of the antigen was determined. The IBDV antigen and the antigen of ND La Sota strain and IB M41 strain prepared by conventional method concentrated and inactivated and the ND-IB-IBD triple inactivated vaccine was prepared and its physical properties were tested,the chickens were immunized with the ND-IB-IBD triple vaccine with different doses to determine its immune effect. The results showed that the virus titer of the IBDV BJQ902 strain could reach 107.78.5 TCID50/0.1 mL after IBDV BJQ902 strain was inoculated with suspended DF-1 cells. The prepared ND-IB-IBD triple vaccine has stable physical properties. Good protection can be obtained by immunizing chickens with 0.1 mL/chicken. The results of this study could provide experimental data for the new vaccines development.
