摘 要:目的探讨DNA条形码技术鉴定本地郊区常见鼠形动物的可行性。方法收集134份本地常见鼠形动物样本,PCR扩增线粒体细胞色素C氧化酶亚基I基因,测序后进行序列比对,并计算物种种内和种间遗传距离,采用邻接法构建系统发育树。结果序列比对结果显示共有黑线姬鼠22只、中华姬鼠19只、小家鼠25只、褐家鼠39只、社鼠19只、白腹鼠未定种4只、山西[鼠平]只、大仓鼠2只、川西缺齿鼩鼱2只。其中,有2份现场形态学鉴定为小家鼠的样本,经分子方法鉴定为褐家鼠,提示现场鉴定有误。系统发育分析显示,各物种种内遗传距离远小于种间遗传距离;所采集样本在进化树上形成8个高度支持的独立分支,与形态学鉴定结果基本一致。结论 DNA条形码技术能有效鉴定本地鼠形动物种类,有助于提高本地鼠形动物鉴定的准确性。Objective To explore the feasibility of DNA barcoding technology in identifying common murine-like species in the suburbs of Beijing.Methods A total of 134 local common murine-like animals samples were collected,and each of mitochondrial cytochrome C oxidase subunit I genes was amplified and sequenced.Sequence alignment was performed and the intraspecific and interspecific genetic distances were analyzed.Phylogenetic tree was constructed by neighbor-jointing method.Results The sequence alignment results showed that there were 22 Apodemus agrarius,19 Apodemus draco,25 Mus musculus,39 Rattus norvegicus,19 Niviventer confucianus,4 Niviventer sp.,2 Myodes shanseius,2 Tscherskia Triton,and 2 Chodsigoa hypsibia.Among them,two samples morphologically identified as Mus musculus were identified as Rattus norvegicus by molecular method finally,suggesting the morphological identification result may be incorrect.Phylogenetic analysis showed that the intraspecific genetic distance of each species was much smaller than the interspecific genetic distance.All samples were clustered into eight highly supported independent branches on the phylogenetic tree,which was consistent with the morphological identification results.Conclusion DNA barcoding technology can effectively identify the local murine-like species and helps to improve the accuracy of identification of local murine-like animals.
