摘  要:目的考察溶液的pH值、磷酸盐缓冲溶液、温度、酸碱盐种类对水溶液中天麻苷元降解速率的影响。方法含量测定的高效液相色谱法(HPLC)条件为流动相乙腈-0.05%磷酸(5∶95,V/V),检测波长为273 nm,柱温35℃,测定天麻苷元在不同条件水溶液中的浓度变化,以剩余药物的百分含量(%)为纵坐标,时间(t)为横坐标,绘制降解动力学曲线,对降解差异显著的条件拟合降解动力学方程。结果天麻苷元在pH 1.2、5.0、11.0及盐酸、碳酸钠、磷酸氢二钠水溶液中稳定性较差,降解反应分别符合一级、零级、三级、零级、零级、三级动力学方程。在pH 4.5、6.8、7.2、9.0水溶液中,浓度为0.01、0.02、0.03、0.04、0.05、0.1、0.2 mol/L的磷酸盐缓冲溶液中,不同温度水溶液中及酒石酸、柠檬酸、氢氧化钠溶液中稳定性较好。结论天麻苷元的水溶液在pH 4.5、6.8、7.2、9.0水溶液,不同浓度的磷酸盐缓冲溶液,不同温度水溶液及酒石酸、柠檬酸、氢氧化钠溶液条件下较稳定。Objective To investigate the effects of pH,phosphate buffer solution,temperature and the types of acids and bases on the degradation rate of gastrodigenin in aqueous solution. Methods The HPLC method was used to determine the concentration of gastrodigenin in aqueous solution under different conditions with acetonitrile-0.05% phosphoric acid(5 ∶ 95,V/V)as mobile phase. The detection wavelength was 273 nm and the column temperature was 35 ℃. The degradation kinetics curve was plotted with the percentage of residual drug(%)as ordinate and time(t)as abscissa. The degradation kinetics equation was fitted to the condition that the degradation difference was significant. Results The stability of gastrodigenin was rather poor in pH 1.2,5.0,11.0,hydrochloric acid solution,sodium carbonate solution,and disodium hydrogen phosphate solution. The gastrodigenin followed first-order,zero-order,thirdorder,zero-order,zero-order and third-order kinetics respectively. The stability of gastrodigenin was good under pH 4.5,6.8,7.2,9.0,phosphate buffer solutions with concentrations of 0.01,0.02,0.03,0.05,0.1 and 0.2 mol/L,different temperatures,tartaric acid solution,citric acid solution and sodium hydroxide solution. Conclusion Gastrodigenin in aqueous solution was stable in pH 4.5,6.8,7.2,9.0,phosphate buffer solutions with different concentrations,different temperatures,tartaric acid solution,citric acid solution and sodium hydroxide solution.
