摘 要:目的考察经鼻给药雷公藤红素对小鼠鼻腔和嗅球的影响以及蛙上颚纤毛的毒性。方法以在体蛙上颚黏膜为模型,研究不同给药剂量的雷公藤红素对纤毛转运速率、持续运动时间(PVD)及黏膜纤毛形态的影响。HE染色观察不同给药剂量下对小鼠鼻腔黏膜及嗅球组织的影响。结果与对照组相比,低剂量组纤毛转运速率和PVD有所下降(P<0.05),黏膜组织相对完好,极少量充血;中剂量转运速率和PVD明显下降(P<0.01),表面纤毛组织脱落,黏膜组织充血明显;高剂量组转运速率接近于0,PVD仅为对照组的4%(P<0.01),纤毛组织极少存留,黏膜组织大量充血,几乎覆盖沟状结构。小鼠鼻腔内黏膜表面炎症浸润及嗅球实变程度与给药剂量呈正相关。结论蛙上颚黏膜给药浓度为0.1 mg/mL的雷公藤红素时,纤毛转运速率、持续运动时间及黏膜纤毛形态无影响;小鼠经鼻给药剂量为2 mg/kg的雷公藤红素时,鼻腔结构和嗅球组织无显著影响。Objective To study the effect of celastrol on the nasal cavity and the olfactory bulb of mice and the toxicity of the cilia in the frog. Methods In vivo, the maxillary mucosa of the frog was used as the model. The effects of different doses of tripterine on cilia transport rate, duration of continuous motion (PVD) and the mucociliary morphology were studied. HE staining was used to observe the effects of different administration doses on the nasal mucosa and the olfactory bulb of mice. Results Compared with the control group, the rate of cilia transport and PVD in low-dose group decreased (P<0.05), and the mucosal tissue was relatively intact, with a very small amount of hyperemia. The transport rate of high-dose group was close to 0 and PVD was only 4% (P<0.01) in the control group, and the ciliated tissues were very rare, and the mucosal tissue was hyperemia and almost covered the groove-like structure. There was a positive correlation between the inflammatory infiltrates of the mucosal surface and the level of the olfactory bulb in the nasal cavity of the mice. Conclusion The rate of cilia transport, the duration of continuous motion and the morphology of the mucoccilia were not affected when the concentration of tripterine was 0.1 mg/mL, and there was no significant effect on the structure of the nasal cavity or the tissue of the olfactory bulb when the mice were given a dose of 2 mg/kg of celastrol.
