摘 要:目的昆明山海棠在自身免疫性疾病中疗效显著,但生殖系统不良反应严重危害患者健康。使用网络药理学方法对昆明山海棠生殖毒性的分子机制进行研究,寻找昆明山海棠减毒配伍具有重要临床价值。方法通过网络药理学方法构建药物-靶点网络和不良反应-靶点网络,进而寻找昆明山海棠可能作用的闭经相关靶点,并与昆明山海棠成分进行映射。结果发现昆明山海棠的10个有效成分可能靶向16个闭经相关靶点,通过台湾三尖杉碱、儿茶酚、表没食子儿茶素、雷酚二萜酸、雷酚酮内酯等靶向雌激素受体、孕激素受体、雄激素受体、肾上腺素能受体等。结论昆明山海棠可能通过对性激素的影响导致卵巢性闭经。本研究基于药物的治疗作用,进一步探究了药物的不良反应,对中药配伍减毒研究提供了一定的思路,具备一定研究创新性和临床价值。Objective Kunming Shanhaitang(Tripterygium Hypoglaucum) is effective in autoimmune diseases,but adverse effects of reproductive system,such as amenorrhea,are seriously harmful to patients′ health.The molecular mechanism of Tripterygium Hypoglaucum on reproductive toxicity was studied by network pharmacology,and it is valuable for clinical application to find out a way to reduce toxicity.Methods The drug-target network and amenorrhea-target network was constructed.Furthermore,the amenorrhea-related targets were found,and mapped with the components of Tripterygium Hypoglaucum.Results Ten components might target 16 proteins and estrogen receptors,progesterone receptors,androgen receptor,adrenergic receptor might be targeted by wilsonine,catechin,epigallocatechin,triptonoditerpenic acid,triptonolide etc..Conclusion Tripterygium Hypoglaucum may cause ovarian amenorrhea by its influence on sex hormones.Based on the therapeutic effect of the Tripterygium Hypoglaucum,we further explored the adverse reactions,which provided a way to study the combination of herbs to reduce the toxicity of traditional Chinese medicine,which meant our researches were of significance and innovation.
