摘  要:目的探讨黄芪甲苷(ASⅣ)及人参皂苷Rg1(Gin Rg1)对高脂大鼠心肌缺血再灌注损伤(MIRI)后心肌细胞凋亡的影响。方法选取40只健康成年雄性SD大鼠随机分为假手术组(S组)、高脂模型组(M组)、黄芪甲苷治疗组(A组)、人参皂苷Rg1治疗组(G组)、黄芪甲苷+人参皂苷Rg1合用治疗组(C组)5组,每组8只。采用高脂膳食喂养的方法复制高脂大鼠模型,再通过可逆性左冠状动脉前降支结扎法建立MIRI大鼠模型,假手术组仅穿线不结扎。药物组大鼠给予相应的药物灌胃,每日3次,疗程1周,假手术组和高脂模型组给予等量的0.9%氯化钠注射液。治疗结束后,取腹主动脉血检测各组eNOS、Cytc、Bax、Bcl-2蛋白的表达水平。结果与假手术组相比,模型组大鼠eNOS、Bcl-2蛋白的相对表达水平显著降低,Cytc、Bax蛋白的相对表达水平显著升高,差异有统计学意义(P<0.01);与模型组相比,药物组大鼠的eNOS、Bcl-2蛋白的相对表达水平升高,Cytc、Bax蛋白的相对表达水平显著下降,差异具有统计学意义(P<0.01或P<0.05)。结论黄芪甲苷及人参皂苷Rg1对高脂大鼠MIRI的保护作用可能是通过抑制Cytc释放、调控凋亡蛋白Bax、Bcl-2的表达来实现的,调控过程与上调eNOS蛋白的表达密切相关。Objective The present study aimed to investigate the effects of astragaloside Ⅵ and ginsenoside Rg1 on myocardial apoptosis after myocardial ischemia reperfusion injury(MIRI) in hyperlipidemia rats.Methods Forty healthy adult male SD rats were randomly divided into 5 groups including the sham operated group(S group),the hyperlipidemic mice model group(M group),the astragaloside Ⅵ treatment group(A group),the ginsenoside Rg1 treatment group(G group) and the combined astragaloside Ⅵ with ginsenoside Rg1 treatment group(C group),with 8 rats in each group.The high-fat diet was used to replicate the hyperlipidemia rat model.The rat model of MIRI was replicated by clamping the coronary artery.The sham operated group was only threaded without ligation.Rats in the drug groups were given corresponding drugs by gavage for 3 times a day for 1 week,while rats in the sham group and model group were given the same amount of 0.9% NaCl.After 1 week,Western blot analysis was used to detect the relative expression levels of eNOS,Cytc,Bax and Bcl-2 protein.Results Compared with the sham operated rats,the expression levels of eNOS and Bcl-2 protein in the model group were decreased and the expression levels of Cytc and Bax protein were increased(P<0.01).Compared with those of the model group,the expression levels of eNOS and Bcl-2 protein were increased and the expression levels of Cytc and Bax protein were decreased in the drug groups(P<0.01 or P<0.05).Conclusion Astragaloside Ⅵ and ginsenoside Rg1 can reduce MIRI in hyperlipidemia rats via inhibiting the release of Cytc to regulate myocardial apoptosis and the regulatory process was closely related to up-regulation of eNOS protein.
