摘 要:目的对1株南海柳珊瑚来源真菌Penicillium chrysogenum(CHNSCLM-0019)中的生物碱类化合物进行研究。方法综合运用正、反相硅胶柱层析、高效液相色谱(HPLC)及重结晶等方法对真菌粗提物进行分离纯化,并通过现代波谱学方法、单晶衍射及核磁共振变温实验并结合文献对所得化合物进行结构鉴定。结果鉴定了6个苯并二氮杂卓类生物碱1~6,分别为7-methoxycyclopenin (1),cyclopenin (2),7-methoxycyclopeptin(3),(3S)-1, 4-benzodiazepine-2, 5-diones (4),7-methoxydehydrocyclopeptin (5),(+)-cyclopenol (6)。并通过核磁共振变温实验验证了化合物1以互变构象异构体形式存在。结论柳珊瑚来源真菌P. chrysogenum(CHNSCLM-0019)是苯并二氮杂卓类生物碱的重要来源。Objective To study the alkaloids from the gorgonian coral-derived fungus Penicillium chrysogenum(CHNSCLM-0019). Methods The pure compounds were isolated by Silica gel column chromatography, ODS, HPLC and recrystallization. Their structures were determined by modern spectroscopic methods, X-ray, variable temperature NMR, and comparison with previously reported data. Results The structures of six benzodiazepine alkaloids 1~6 were determined as 7-methoxycyclopenin(1), cyclopenin(2), 7-methoxycyclopeptin(3),(3 S)-1, 4-benzodiazepine-2, 5-diones(4), 7-methoxydehydrocyclopeptin(5), and(+)-cyclopenol(6). Compound 1 was determined as tautomorphic isomers. Conclusion The gorgonian coralderived fungus P. chrysogenum was an important resource of benzodiazepine alkaloids.
