摘 要:目的观察柴胡疏肝散对抑郁症模型大鼠SIRT1/NF-κB信号通路的作用,探讨柴胡疏肝散抗抑郁可能机制。方法将大鼠随机分为空白组、模型组、西酞普兰组和柴胡疏肝散组,除空白组外,其他各组制备抑郁症模型。西酞普兰组给予草酸艾司西酞普兰片制成的药液柴胡疏肝散组给予柴胡疏肝散药液,空白组和模型组给予等体积的纯水。采用旷场实验、强迫游泳实验和蔗糖偏好实验用于评价柴胡疏肝散的抗抑郁作用。ELISA检测大鼠血清中TNF-α、IL-1β的表达水平。免疫组化和Western bolt检测前额叶皮层中SIRT1、NF-κB、IκBα、Caspase3的蛋白表达水平。结果与空白组比较,模型组大鼠糖水偏好率、运动距离和穿越中央区域次数显著减少(P<0.01),而强迫游泳不动时间和静止时间显著增加(P<0.01)。与模型组比较,西酞普兰组和柴胡疏肝散组大鼠糖水偏好率、运动距离和穿越中央区域次数增加(P<0.01,P<0.05),而强迫游泳不动时间和静止时间均有不同程度减少(P<0.01,P<0.05)。与空白组比较模型组大鼠血清中TNF-α、IL-1β表达水平明显升高(P<0.01);与模型组比较,西酞普兰组和柴胡疏肝散组大鼠血清中TNF-α、IL-1β表达水平降低(P<0.01,P<0.05)。与空白组比较,模型组大鼠前额叶皮层中SIRT1、IκBα的IOD值和蛋白水平明显降低(P<0.01),而NF-κB、Caspase3的IOD值和蛋白水平明显升高(P<0.01)。与模型组比较,西酞普兰组和柴胡疏肝散组大鼠前额叶皮层中SIRT1、IκBα的IOD值和蛋白水平升高(P<0.01,P<0.05),而NF-κB、Caspase3的IOD值和蛋白水平降低(P<0.01,P<0.05)。结论柴胡疏肝散具有抗抑郁作用,其机制可能与其调节SIRT1/NF-κB信号通路有关。Objective To observe the effects of Chaihu Shugan Powder on SIRT1/NF-kappa B signaling pathway in depression model rats,and explore the possible anti-depression mechanism of Chaihu Shugan Powder.Methods Rats were randomly divided into blank group,model group,citalopram group and Chaihu Shugan Powder group.Besides blank group,depression models were established in other groups.Citalopram group was given Escitalopram Oxalate Tablets,Chaihu Shugan Powder group was given Chaihu Shugan Powder,blank group and model group were given equal volume of pure water.The open field experiment,forced swimming experiment and sucrose preference experiment were used to evaluate the antidepressant effects of Chaihu Shugan Powder.The expression levels of TNF-α and IL-1β in serum of rats were detected by ELISA.The expression levels of SIRT1,NF-κB,I-κBα and Caspase-3 in prefrontal cortex were detected by immunohistochemistry and Western blot.Results Compared with blank group,the sugar preference rate exercise distance and times of crossing the central area of model group were significantly increased(P<0.01),while the immobility time and rest time of forced swimming were significantly reduced(P<0.01);compared with model group,the preference rate of sugar and water,the distance of exercise and the number of times of crossing the central area of rats in citalopram group and Chaihu Shugan Powder group decreased(P<0.01,P<0.05),while the immobility time and static time of forced swimming increased in varying degrees(P<0.01,P<0.05).Compared with blank group,the serum levels of TNF-α and IL-1β in model group were significantly higher(P<0.01);compared with model group,the levels of TNF-α and IL-1β in serum of rats in citalopram group and Chaihu Shugan Powder group decreased(P<0.01,P<0.05);compared with blank group,the IOD values and protein levels of SIRT1 and I-κBα in the prefrontal cortex of model group were significantly lower(P<0.01),while the IOD values and protein levels of NF-κB and Caspase-3 were significantly higher(P<0.01);
