摘 要:新疆野苹果是苹果属重要的抗逆育种资源,观赏价值较高,是园林绿化的优良树种。以18个新疆野苹果无性系为研究对象,基于SSR分子标记技术对其进行分子鉴定,并构建了DNA指纹图谱。结果表明:13对SSR引物共扩增出62条条带,每对引物扩增条带数在3~6条之间,平均每对引物扩增条带4.77条,每对引物的多态位点百分比为100%,各引物的多态性信息含量PIC值的变化范围为0.512~0.879。在遗传距离0.566处,18个新疆野苹果无性系被分成4类,第一类为M1、M14、M13、M16、M3、M4、M6等7个无性系;第二类为M151个无性系;第三类为M2、M5、M9、M11、M12等5个无性系;第四类为M7、M17、M18、M8、M10等5个无性系。基于SSR扩增结果,构建18个新疆野苹果无性系的DNA指纹图谱及二维码。Xinjiang wild apple is an important stress-resistant breeding resource of Malus with has high ornamental value and is an excellent tree species for landscape greening.In this study,18 Xinjiang wild apple clones were identified by SSR molecular marker technology,and the DNA fingerprint was constructed.The results showed that a total of 62 bands were amplified by 13 pairs of SSR primers.The number of bands amplified by each pair of primers ranged from 3 to 6.The average number of bands amplified by each pair of primers was 4.77.The percentage of polymorphic loci per pair of primers was 100%.The PIC value of polymorphic information content of each pair of primers ranged from 0.512 to 0.879.18 Xinjiang wild apple clones were classified into four groups at genetic distance of 0.566.The first category is 7 clones:M-1,M-14,M-13,M-16,M-3,M-4,M-6;the second category is 1 clone:M-15;the third category is 5 clones:M-2,M-5,M-9,M-11,M-12;the fourth category is 5 clones:M-7,M-17,M-18,M-8,M-10.Based on the results of SSR amplification,DNA fingerprints and two-dimensional codes of 18 wild apple clones were constructed.
