摘  要:目的:探讨加味温胆汤对抑郁模型大鼠神经营养因子及前额叶皮质神经元损伤的影响,为其发挥抗抑郁作用提供实验依据。方法:27只SD大鼠随机分为正常对照组、模型组、中药组,每组9只。采用孤养结合慢性不可预见性温和应激(CUMS)法复制抑郁大鼠模型,连续21天造模成功后,模型组给予生理盐水2 mL/(kg·d),中药组给予加味温胆汤12 g/(kg·d),各组均灌胃给药,连续28日。采用旷场实验和糖水偏好实验检测加味温胆汤的抗抑郁效应,HE染色观察前额叶皮质形态结构变化,Elisa法检测各组大鼠血清脑源性神经营养因子(BDNF)、胶质细胞源性的神经营养因子(GDNF)、转化生长因子-β(TGF-β)含量变化。结果:与正常对照组比较,抑郁模型组大鼠旷场实验得分、糖水偏好率低于正常对照组,差异有统计学意义(P<0.01);与模型组比较,加味温胆汤组得分高于模型组,差异有统计学意义(P<0.01)。HE染色结果显示,与正常对照组相比,模型组大鼠前额叶皮质区域神经元胞体肿胀明显,细胞排列紊乱,细胞核固缩,细胞质染色加深,脑区损伤情况较为严重;在给予加味温胆汤干预后,模型大鼠前额叶皮质神经元损伤明显缓解,结构基本正常,脑区损伤情况得到显著缓解。Elisa检测结果显示,与正常对照组相比,模型组血清BDNF、GDNF和TGF-β水平下降,差异有统计学意义(P<0.01);与模型组相比,加味温胆汤组血清BDNF、GDNF和TGF-β水平上升,差异有统计学意义(P<0.05或P<0.01)。结论:加味温胆汤可改善模型大鼠的抑郁行为,其作用机制可能与提高血清神经营养因子BDNF、GDNF和TGF-β水平,修复前额叶皮质损伤有关。Objective:To investigate the effects of Jiawei Wendan Decoction on neurotrophic factors and neurons injury of prefrontal cortex in depression model rats,so as to provide experimental basis for its antidepressant effect.Methods:27 SD rats were randomly divided into the normal control group,the model group and the Chinese medication group,with 9 rats in each group.The depression model was established by the method of isolation combined with chronic unpredictable mild stress(CUMS).After 21 day's of modeling,the model group was given normal saline(2 mL/kg)and the Chinese medication group was treated with Jiawei Wendan Decoction(12 g/kg),After 28 days of intervention,the antidepressant effect of Jiawei Wendan Decoction was detected by open field test and sucrose preference test.HE staining was used to observe the changes of the morphology and structure of prefrontal cortex.Elisa method was used to detect the changes of serum neurotrophic factors,such as BDNF,TGF-βand GDNF,in the rats of each group.Results:The open field test score and sucrose preference rate were significantly lower in the model group than those in the normal group after the intervention(P<0.01);the score and the rate were significantly higher after the treatment in the Jiawei Wendan Decoction group compared to those in the model group(P<0.01).HE staining results showed that in the model group,the neuron body of the prefrontal cortex was obviously swollen,the cell arrangement was in disorder,with karyopyknosis and deep cytoplasmic staining and severe brain injury.After the intervention of Jiawei Wendan Decoction,the damage of neurons of the prefrontal cortex was obviously alleviated,the structure was basically normal and the damage of brain was significantly alleviated.Elisa test results showed that the serum levels of BDNF,GDNF and TGF-βdecreased significantly in the model group compared to those in the normal group(P<0.01);the serum levels of BDNF,GDNF and TGF-βincreased significantly in the Jiawei Wendan Decoction group after the treatment compa
