摘 要:目的拟探讨基于叶绿体基因片段psbA-trnH与matK序列的DNA条形码技术在淫羊藿属中的分类与鉴定中的可行性。方法选用不同来源的34个种,共计84份材料,对其psbA-trnH与matK序列片段进行PCR扩增、测序,分析其序列组成和结构特点,应用Kimura双因素模型(Kimura-2-parameter,K-2-P)计算遗传距离,同时应用邻接法(Neighbor-Joining,N-J)构建系统发育树。结果PsbA-trnH序列在34种淫羊藿属材料中的通用性较matK好;通过K-2-P模型评估,psbA-trnH鉴定成功率为36.7%,matK鉴定成功率为24.2%,2条序列遗传距离均存在一定gap;N-J进化树显示,matK序列N-J系统进化树鉴定效率为17.1%,psbA-trnH序列N-J系统进化树鉴定效率为25.7%。结论本研究丰富了淫羊藿属植物的分类鉴定资料,但在淫羊藿属中的分类与鉴定中,应用基于叶绿体基因片段psbA-trnH与matK序列的DNA条形码技术仍具有一定局限性。Objective This study intends to explore the feasibility of taxonomy and identification in Epimedium(Berberidacae)applying DNA Barcoding technology based on chloroplast gene fragments psbA-trnH and matK sequences.Methods A total of 84 materials from 34 species of different sources were selected to amplify and sequence the psbA-trnH and matK sequence fragments by PCR.Their sequence composition and structure were analyzed.The genetic distance was calculated by Kimura two-factor model(K-2-P),and the phylogenetic tree was constructed by Neighbor-Joining(N-J).Results The results showed that the generality of PsbA-trnH sequence in 34 species of Epimedium was better than that of matK.According to the K-2-P model,the identification success rate of psbA-trnH was 36.7%,that of matK was 24.2%,and there were some gaps between the two sequences.The N-J phylogenetic tree showed that the identification efficiency of matK sequence was 17.1%,and that of psbA-trnH sequence was 25.7%.Conclusion It indicated that this study enriched the taxonomic identification data of Epimedium,but applying DNA barcoding based on chloroplast gene fragments psbA-trnH and matK sequences in taxonomy and identification in Epimedium(Berberidacae)in this study,was quite limited.
