肝宁方对肝硬化大鼠炎症因子和氧化应激的影响及机制研究

摘　　要：目的探讨肝宁方对肝硬化模型大鼠炎症因子和氧化应激的影响以及可能机制研究。方法55只大鼠随机取10只作为对照组,剩余45只大鼠采用四氯化碳的复合因素法进行肝硬化造模。造模8周末,造模组大鼠死亡9只,随机取3只大鼠验证造模成功。正常组大鼠10只,造模组大鼠随机平均分为模型组、肝宁方组、秋水仙碱组,每组11只。对照组和模型组给予生理盐水、肝宁方组给予中药液灌胃,均为10 mL·kg^(-1)·d^(-1);秋水仙碱组给予秋水仙碱0.1 mg·kg^(-1)·d^(-1),共6周。检测大鼠血清肝功能ALT、AST、ALB和TBIL水平;检测大鼠血清肝纤维化四项HA、III-PC、IV-C和LN含量;检测肝组织SOD、GSH-PX和MDA活性;检测大鼠血清IL-1β、IL-6和TNF-α含量;检测大鼠血浆内毒素含量;检测TLR4、NF-κB和MyD88 mRNA及其蛋白表达。结果与模型组比较,肝宁方组大鼠肝功能ALT、AST和TBIL显著降低,ALB显著升高;肝宁方组大鼠肝纤维化四项HA、III-PC、IV-C和LN含量显著降低;肝宁方组大鼠SOD和GSH-PX活性显著增强,MDA活性显著降低;肝宁方组大鼠IL-1β、IL-6、TNF-α和内毒素含量显著降低;肝宁方组大鼠TLR4、NF-κB和MyD88 mRNA及其蛋白表达量显著降低。结论肝宁方可能通过干预肝硬化大鼠肝组织TLR4/NF-κB信号通路的表达,发挥抑制炎症反应、抗氧化应激作用,从而保护肝细胞、抗肝纤维化,延缓肝硬化进展。Objective To study the effect of Ganning Formula(肝宁方)on inflammatory factors and oxidative stress in cirrhotic rats and its possible mechanism.Methods Fifty-five rats were randomly selected 10 rats as the control group,the remaining 45 rats were set up the liver cirrhosis model by carbon tetrachloride complex factor method.At the end of 8 weeks,9 rats died and 3 rats were randomly selected to verify the success of the model.Ten rats in the normal group were randomly divided into model group,Ganning Formula group and colchicine group.The control group and model group were given normal saline and Ganning Formula group were given traditional Chinese medicine solution by gavage,both of which were 10 mL·kg^(-1)·d^(-1).The colchicine group was given colchicine 0.1 mg·kg^(-1)·d^(-1) for 6 weeks.The serum levels of ALT,AST,ALB and TBIL were measured.The contents of HA,III-PC,IV-C and LN in serum of rats with hepatic fibrosis were measured.The activities of SOD,GSH-Px and MDA in liver tissue were detected.The levels of IL-1β,IL-6 and TNF-αin serum were measured.The content of endotoxin in plasma was measured.TLR4,NF-κB and MyD88 mRNA and protein expressions were detected.Results After experiment,compared with those of the model group,the levels of ALT,AST and TBIL of liver function in Ganning Formula group were significantly decreased,but the level of ALB was significantly increased.The contents of HA,III-PC,IV-C and LN in liver fibrosis of rats in Ganning Formula group were significantly reduced.The activity of SOD and GSH-Px in Ganning Formula group was significantly increased,and the activity of MDA was significantly decreased.The contents of IL-1β,IL-6,TNF-αand endotoxin in Ganning Formula group were significantly decreased.The expressions of TLR4,NF-κB and MyD88 mRNA and protein decreased significantly in Ganning Formula group.Conclusion Ganning Formula may play the role of inhibiting inflammatory reaction and anti oxidative stress by interfering the expression of TLR4/NF-κB signal pathway in liver t
