摘  要:大多数神经退行性疾病与神经元中聚集性蛋白在胞浆中的沉积和线粒体功能障碍有关,而自噬是去除这些蛋白质和维持线粒体稳态的强有力的过程。自噬是一种高度保守的分解代谢过程,它将细胞质内容物传递给溶酶体以降解,不仅能够清除受损的蛋白质和线粒体,而且使细胞能量循环再利用。总结了自噬的基本过程、相关分子机制、信号途径以及自噬功能障碍与神经退行性疾病的关系,并且初步分析中药对神经细胞自噬的影响,探讨中药有效成分或中药复方通过调节自噬治疗神经退行性疾病的潜在可能。Most neurodegenerative diseases that afflict humans are associated with the intracytoplasmic deposition of aggregate-prone proteins in neurons and mitochondrial dysfunction.Autophagy is a powerful process for removing such proteins and maintaining mitochondrial homeostasis.Autophagy is a conserved pathway that delivers cytoplasmic contents to the lysosome for degradation,which not only clears damaged proteins and mitochondria,but also recycles and preserves cellular energy.In this review,we summarized the basic processes of autophagy,related molecular mechanisms,signaling pathways and the relationship between autophagy dysfunction and neurodegenerative diseases,analyzed the effect of traditional Chinese medicine on autophagy of nerve cells and explored the potential effect of active ingredients or compound formula of traditional Chinese medicine to treat neurodegenerative diseases by regulating autophagy.
