摘  要:为了探究彩色马铃薯薯肉颜色分布的机理,本试验利用RT-PCR技术,检测了与花青素合成相关的6种结构基因和4种调节基因,在彩色马铃薯杂交组合后代中不同薯肉颜色个体以及同一薯块中不同薯肉颜色部分的表达情况,研究这些结构基因和调节基因在薯肉中的表达模式。结果显示,在75个杂交后代中,除结构基因DFR外,其余5个结构基因在红色和紫色薯肉中都有较高的表达量,而在81%(StCHS)~93%(StF3H)黄色或白色薯肉中表达量比较低甚至不表达;4个调节基因在90%(StAN2)~100%(StMBA113)黄色或白色薯肉以及所有紫色(除G17-23-36外)和红色薯肉中都有表达。本研究结果表明,来自同一父母本的后代薯肉颜色的差异可能是由结构基因表达量的差异或者在花青素合成通路存在结构基因没有表达而导致,而且4个调节基因可能并不是限制花青素合成的关键转录因子。In order to explore the mechanism of color expression in the tuber flesh of colorful potato, in this study,6 structural genes and 4 regulatory genes related to anthocyanin synthesis were detected by RT-PCR. The expression patterns of these structural genes and regulatory genes in the tuber flesh were studied by detecting samples which were the offspring of a color potato hybrid cross. Among the 75 offsprings, except for the structural gene DFR, the remaining 5 structural genes had higher expression levels in red and purple flesh, while the expression levels in 81%(StCHS)~93%(StF3 H) yellow or white flesh portion were lower or even not expressed. Four regulatory genes were expressed in 90%(StAN2) to 100%(StMBA113) yellow or white flesh portion and all purple(except G17-23-36) and red flesh portion. The results showed that the differences in the color of flesh in the offspring of the same parent may be caused by the differences in the expression level of structural genes or the lack of expression of structural genes in some steps of anthocyanin synthesis pathway. Moreover, the regulatory genes in this study may not be the key transcription factors limiting anthocyanin synthesis.
