摘 要:本研究利用筛选出的17对多态性高、条带清晰的SSR引物,以来源不同的32份黄瓜栽培品种为材料,进行遗传多样性分析和指纹图谱的构建。结果表明:筛选出的17对多态性引物共检测出78个基因位点,每个引物平均4.59个,其中,多态性扩增位点56个,多态性比例71.8%,引物多态性信息含量(polymorphism information content, PIC)平均值为0.62,变化范围为0.30~0.87;32份黄瓜栽培品种的遗传相似系数(genetic similarity, GS)介于0.560~0.947之间,平均值为0.797,采用UPGMA法进行聚类,在遗传相似系数(GS)为0.760处可将32份黄瓜品种聚为5类;利用这17对SSR引物构建的指纹图谱能够有效区分所有供试材料,并且为每一份材料建立了一份独特的DNA指纹图谱,该结果可为今后黄瓜新品种选育及品种鉴定提供重要依据。The SSR genetic diversity and fingerprint map of 32 cucumber cultivars from different sources were studied using 17 pairs of selected polymorphic SSR primers. The results indicated that 78 loci were detected with a mean of 4.59, and 56 loci were polymorphic with a ratio of 71.8%, by 17 screened polymorphic SSR primers. The polymorphism information content(PIC) varied from 0.30 to 0.87 with an average of 0.62. The genetic similarity coefficient(GS) of 32 cucumber cultivars ranged from 0.560 to 0.947, with an average of 0.797. 32 cucumber cultivars were divided into 5 groups at the genetic similarity coefficient(GS) of 0.760 by UPGMA cluster analysis. The fingerprint data of all the 17 pair of SSR primers were employed to establish the fingerprint database to distinguish the 32 samples and every accession had its unique fingerprint. These results can provide important basis for variety breeding and cucumber identification in the future.
