摘　　要：通过笔筒树孢子的无菌培养试验,以筛选出笔筒树孢子无菌培养各过程的最佳条件,建立一套系统、高效、完整的笔筒树的无菌培养技术体系,最终实现笔筒树种苗的工厂化生产。本研究用即采的成熟的离叶黑褐色孢子为材料,用75%酒精处理10 s后再用0.1%升汞处理6 min结合进行笔筒树孢子的表面消毒,诱导孢子萌发的最适培养基为1/2 MS+6-BA 0.1 mg/L+IBA 0.2 mg/L,增殖培养基为MS+6-BA 0.05 mg/L+IBA 0.05 mg/L,分化培养基为MS+6-BA 0.2 mg/L+IBA 0.2 mg/L,生根培养基为MS+6-BA 0.05 mg/L+IBA0.05 mg/L,培养温度(25±2)℃,日光灯光源,光照时间12 h/d,光照强度2 000 lx。Through the aseptic culture test of Sphaeropteris lepifera(J Sm. ex Hook.) R. M. Tryon, try to screen out the best conditions for each process of aseptic culture of pen spores, in order to establish a systematic, efficient and complete, aseptic culture technology system of pen spores, and try to achieve factory production of pen barrel tree seedlings. The mature black and brown spores separated from blades of S. lepifera were used as explants,disinfected with 75% alcohol for 10 s and 0.1% Hg for 6 min. The induced germination medium is 1/5 MS+6-BA0.5 mg/L+IBA 0.5 mg/L. The enrichment medium is 1/2 MS+6-BA 0.3 mg/L+IBA 0.05 mg/L. The differential medium is MS+6-BA 0.05 mg/L+IBA 0.05 mg/L. The culture temperature is(25±2) ℃, the light source is fluorescent,the illumination time is 12 h/d, and the illumination intensity is 2 000 lx.
