摘 要:目的:观察鸡血莲黄酮对非酒精性脂肪肝(NAFLD)小鼠的保护作用及机制。方法:40只雄性昆明小鼠随机分为正常组、模型组、鸡血莲黄酮高剂量组和鸡血莲黄酮低剂量组。除正常组外,其余各组连续饲喂高脂饲料4周制备小鼠NAFLD模型,并从第5周开始,鸡血莲黄酮高、低剂量组分别以200、100 mg/kg的鸡血莲黄酮连续灌胃4周,最后一次灌胃后,各组小鼠禁食12 h,取血,取肝脏。试剂盒检测小鼠血清AST、ALT水平、血脂水平和肝组织抗氧化酶活性;ELISA试剂盒检测肝脏炎症因子水平。HE染色观察肝脏的病理改变,Western blotting检测肝组织NF-κB、AMPK和Sirt1的表达。结果:与模型组比较,鸡血莲黄酮能明显降低小鼠血清TC、TG、AST和ALT水平,降低肝组织中MDA含量,增加CAT、SOD及GSH-Px的含量,减轻小鼠肝细胞脂肪变性及炎性浸润,降低TNF-α、IL-1β和IL-6含量,下调肝组织NF-κB的表达,上调AMPK及Sirt1的表达。结论:鸡血莲黄酮通过调控AMPK/Sirt1/NF-κB通路改善NAFLD小鼠脂代谢紊乱,降低炎症反应及氧化应激。Objective: To observe the protective effect and mechanism of flavone from Jixuelian(Pronephrium penangianum) on non-alcoholic fatty liver disease(NAFLD) in mice. Methods: A total of 40 male Kunming mice were randomly divided into normal group, model group, flavone of Jixuelian(Pronephrium penangianum) high-dose group and flavone of Jixuelian(Pronephrium penangianum) low-dose group. Except for the normal group,the other groups were fed with high-fat diet for 4 weeks. From the 5 thweek, flavone of Jixuelian(Pronephrium penangianum) high, low-dose group was given 200 or 100 mg/kg flavone of Jixuelian(Pronephrium penangianum)by gavage. After the last gavage, mice in each group fasted for 12 hours, and blood and liver were taken. The serum AST, ALT levels, blood lipid levels and liver tissue antioxidant enzyme activity were detected by kits.The liver pathology was observed by HE staining, and the expressions of NF-κB, AMPK and Sirt1 in liver tissues were detected. Results: Compared with the model group, flavone of Jixuelian(Pronephrium penangianum) can significantly reduce the levels of serum TC, TG, AST and ALT in mice, reduce the content of MDA in liver tissue, increase the content of CAT, SOD and GSH-Px in liver tissue, reduce the fatty degeneration and inflammatory infiltration of mouse liver cells, reduce the content of TNF-α, IL-1β and IL-6, down-regulate the expression of liver tissue NF-κB, and up-regulate the expression of AMPK and Sirt1. Conclusion: The flavone of Jixuelian(Pronephrium penangianum) can improve the lipid metabolism disorder of NAFLD mice by regulating the AMPK/Sirt1/NF-κB pathway, and reduce the inflammation and oxidative stress.
