摘　　要：目的研究通关藤提取物(Marsdenia tenacissima extract,MTE)对HepG2肝癌细胞的抑制作用及其机制。方法用MTT法及克隆形成实验检测不同浓度的MTE对HepG2肝癌细胞的增殖抑制作用,并运用流式细胞仪检测不同浓度组24 h后细胞周期的影响。结果不同浓度的MTE对HepG2肝癌细胞均有抑制作用,且呈一定的浓度依赖性及时间依赖性。随着浓度升高,HepG2肝癌细胞G1期细胞比率增加,S期细胞比率降低,与空白对照组比较,差异有统计学意义(P <0.05)。结论 MTE对HepG2肝癌细胞增殖具有明显抑制作用,其作用主要通过干扰HepG2肝癌细胞周期进程,阻滞于G1/S期实现。Objective To study the inhibitory effects and mechanism of Marsdenia tenacissima extract on HepG2 hepatocellular carcinoma cells. Methods The inhibitory effect of different doses of Marsdenia tenacissima extract on the proliferation of hepatocellular carcinoma cell line HepG2 was detected by MTT assay and clone formation experiment. Flow Cytometry detected HepG2 Hepatocellular Carcinoma Cell Cycle at 24 h in each concentration group. Results Different concentrations of Marsdenia tenacissima extract had inhibitory effects on HepG2 Hepatocellular carcinoma cells,and there were concentration-dependent manner and time-dependent manner. With increasing concentrations,the ratio of HepG2 hepatocellular carcinoma cells in G1 phase increased,and the ratio of S phase cells decreased significantly,which was statistically significant compared with the blank control( P < 0. 05). Conclusion Marsdenia tenacissima extract can inhibit the proliferation of HepG2 hepatocellular carcinoma cells,which is mainly achieved by interfering with HepG2 hepatoma cell cycle and blocking in G1 phase.
