摘 要:目的考察menaquinone-7(MK-7)对糖皮质激素致大鼠骨质疏松的作用。方法采用糖皮质激素诱导大鼠骨质疏松,测定血清碱性磷酸酶(alkaline phosphatase,ALP)水平评定造模结果,将造模成功的大鼠随机分组给药。检测骨形态、生物力学指标及相关生化学指标;测定血清骨钙素(bone gla protein,BGP)及钙调蛋白(calmodulin,CAM)水平。结果MK-7能改善模型大鼠的骨小梁结构,显著增加模型大鼠的骨重、骨强度、骨钙、血钙和血清羟脯氨酸(hydroxyproline,Hyp)水平,降低模型大鼠的尿钙和血清ALP水平,并且提升模型大鼠的血清BGP及CAM水平。结论MK-7可以上调血清BGP及CAM水平,增加骨钙沉积及机体钙储备,从而改善糖皮质激素性骨质疏松大鼠的骨形态及骨强度。Objective To investigate the role of MK-7 on glucocorticoid-induced osteoporosis in rats.Methods Osteoporosis model was established with dexamethasone in rats.Bone morphology,biomechanics and biochemical indexes were measured,and the levels of serum bone gla protein(BGP)and calmodulin(CAM)were determined by radioimmunoassay.Results Structure of bone trabecula in MK-7 groups was improved compared with model group.Bone weight,bone strength,the level of serum hydroxyproline(Hyp),the content of bone calcium and serum calcium were increased,while the level of serum alkaline phosphatase(ALP),urinary calcium were decreased by MK-7 in glucocorticoid-treated rats.The level of BGP and CAM in MK-7 groups were increased than those in model group.Conclusion MK-7 could inhibit osteoporosis in glucocorticoid-treated rats through increasing the level of serum BGP and CAM.
