冬凌草素通过抑制NLRP3炎症小体形成减轻蛛网膜下腔出血后的早期脑损伤

摘　　要：目的本研究探讨冬凌草素对蛛网膜下腔出血后NLRP3炎症小体形成的影响。方法采用蛛网膜下腔出血模型,检测蛛网膜下腔出血后神经功能评分和脑水肿情况;Western blot检测NLRP3炎症小体相关蛋白及凋亡相关蛋白的表达情况;TUNEL染色考察神经元凋亡的发生;免疫荧光方法检测小胶质细胞的激活情况。结果冬凌草素能够显著改善神经功能和脑水肿的发生;并且能够有效的抑制NLRP3炎症小体的形成,抑制神经元细胞的凋亡。结论冬凌草素能够抑制NLRP3炎症小体的形成和神经元细胞凋亡,改善蛛网膜下腔出血后的早期脑损伤。Objective To investigate the effect of oridonin on the formation of NLRP3 inflammasome after subarachnoid hemorrhage.Methods Subarachnoid hemorrhage model was used to detect neurological function score and cerebral edema after subarachnoid hemorrhage.Western blot was used to detect the expression of NLRP3 inflammasome related protein and apoptosis-related protein.TUNEL staining was used to investigate the occurrence of neuronal apoptosis.Immunofluorescence method was used to detect the activation of microglia.Results Oridonin could significantly improve neurological function and cerebral edema.Oridonin could effectively inhibit the formation of NLRP3 inflammasome and inhibit the apoptosis of neuron cells.Conclusion Oridonin can inhibit the formation of NLRP3 inflammatory corpuscles and neuronal apoptosis,and improve early brain injury after subarachnoid hemorrhage.
