摘  要:为了研究N-乙酰-β-D-氨基葡萄糖苷酶(NAGase)抑制剂乙酰胺对杜洛克公猪精子顶体完整性及线粒体活性的影响,并初步探讨乙酰胺与精子活力及受精率的相关性及其调控机制,试验通过在精子悬液中加入不同浓度(15,50,100,200 mmol/L)乙酰胺溶液孵育15,30,60 min,采用IVOSⅡCACA计算机辅助精子分析系统和激光共聚焦显微镜检测精子顶体完整率及精子线粒体活性。结果表明:经不同浓度乙酰胺处理后在不同孵育时间下,不同浓度乙酰胺处理的猪精液精子顶体完整率与对照相比均差异极显著(P<0.01);体外孵育15 min时,15,50,100 mmol/L乙酰胺浓度间比较,精子顶体完整率差异不显著(P>0.05);体外孵育30 min时,200 mmol/L乙酰胺处理的猪精液精子顶体完整率极显著低于对照(P<0.01);体外孵育60 min时,50,100,200 mmol/L乙酰胺处理的猪精液精子顶体完整率差异不显著(P>0.05)。体外孵育15,30,60 min时,50,100,200 mmol/L乙酰胺处理的猪精液精子线粒体活性均极显著低于对照(P<0.01)。说明乙酰胺对精子顶体完整性和线粒体活性具有重要的影响。In order to study the effect of N-acetyl-beta-D-amino glycosidase enzymes(NAGase) inhibitor effector acetamideon on the acrosome integrity and mitochondrial activity of Duroc boars sperm, and to explore the correlation and mechanism between acetamide and sperm motility and fertilization rate, different concentrations(15,50,100,200 mmol/L) acetamide solution were added in sperm suspension, and incubated for 15,30,60 min.Using IVOS Ⅱ CACA computer assisted sperm analysis system and laser confocal microscope to analyze the sperm acrosome integrity and mitochondrial activity. The results showed that after treatment with different concentrations of acetamide at different time, the acrosomal integrity rate of sperm was significantly different from that of control(P<0.01).There was no significant difference in acrosomal integrity among 15, 50 and 100 mmol/L acetamide concentrations after in vitro incubation for 15 min(P>0.05).After in vitro incubation for 30 min, the acrosomal integrity rate of 200 mmol/L acetamide treatment was significantly lower than that of control(P<0.01).After 60 min in vitro incubation, there was no significant difference in acrosomal integrity rate of 50, 100 and 200 mmol/L acetamide(P>0.05). After in vitro incubation for 15, 30 and 60 min, the mitochondrial activity of pig semen and sperm treated with 50, 100 and 200 mmol/L acetamide was significantly lower than that of control(P<0.01).The results indicated that acetamide has an important effect on sperm acrosome integrity and mitochondrial activity.
