摘  要:为了解2018年福建省规模化猪场妊娠母猪频繁发生流产、死胎的原因,试验采集福建省30个规模化猪场送检的流产死胎、血清及精液共230份病料,采用RT-PCR和PCR方法对引起母猪繁殖障碍的主要病原猪瘟病毒(CSFV)、猪繁殖与呼吸综合征病毒(PRRSV)、乙型脑炎病毒(JEV)、猪伪狂犬病病毒(PRV)、猪细小病毒(PPV)、猪圆环病毒2型(PCV-2)及猪圆环病毒3型(PCV-3)进行检测和统计。同时对猪场主要病原的目的基因进行克隆、测序,利用分子生物学软件DNAStar 7.0对测序所得序列进行比对分析,利用MEGA6.0软件进行遗传进化分析。结果表明:30个猪场中PRRSV检出率为76.7%(23/30),PCV-2检出率为60.0%(18/30),PRV检出率为43.3%(13/30),PCV-3检出率为10.0%(3/30),其中PCV-2+PRRSV混合感染的猪场检出率为23.3%(7/30),PCV-2+PRRSV+PRV混合感染的猪场检出率为20.0%(6/30),PRRSV+PRV混合感染的猪场检出率为10.0%(3/30),PRRSV+PCV-2+CSFV混合感染的猪场检出率为3.3%(1/30)。23个猪场PRRSV ORF5基因之间的核苷酸相似性为95.1%~99.9%,与JXA1、CH-1a、NADC30、FJFS等参考毒株核苷酸相似性为92.1%~99.6%;18个猪场PCV-2 ORF2基因之间的核苷酸相似性为99.2%~99.9%,与BDH、AF055394、AF055392、Zhuji2003等参考毒株核苷酸相似性为99.2%~99.6%;13个猪场PRV gE基因之间的核苷酸相似性为99.2%~99.8%,与HBCX2013、NYXY、Ea等参考毒株核苷酸相似性为98.3%~99.7%。遗传进化分析结果显示,NADC30-like PRRSV、PCV-2d、PRV变异株成为猪场的优势毒株。说明引起规模化猪场母猪发生繁殖障碍性疾病的主要病原为PRRSV、PRV与PCV-2。In order to understand the causes of frequent miscarriages and stillbirths in pregnant sows on large-scale pig farms in Fujian Province in 2018,230 miscarriage stillbirth,serum and semen samples from 30 large-scale pig farms in Fujian Province were collected in this experiment.RT-PCR and PCR technology were used to detect the major pathogens of Classical swine fever virus(CSFV),Porcine reproductive and respiratory syndrome virus(PRRSV),Japanese encephalitis virus(JEV),porcine Pseudorabies virus(PRV),Porcine parvovirus(PPV),Porcine circovirus type 2(PCV-2)and Porcine circovirus type 3(PCV-3),that cause reproductive disorders in sows,and the data were analysed statistically.At the same time,the target genes of the main pathogens in pig farm were cloned and sequenced,the molecular biology software DNAStar 7.0 was used to alignment and analyze the obtained sequences,and MEGA6.0 software was used for genetic variation analysis.The results showed that the detection rate of PRRSV in 30 farms was 76.7%(23/30),the detection rate of PCV-2 was 60.0%(18/30),the detection rate of PRV was 43.3%(13/30),and the detection rate of PCV-3 was 10.0%(3/30);among them,the infection rate in mixed-infected pig farm of PCV-2+PRRSV was 23.3%(7/30);the infection rate in mixed-infected pig farm of PCV-2+PRRSV+PRV was 20.0%(6/30);the infection rate in mixed-infected pig farm of PRRSV+PRV was 10.0%(3/30);the infection rate in mixed-infected pig farm of PRRSV+PCV-2+CSFV was 3.3%(1/30).The nucleotide similarity among the PRRSV ORF5 genes of 23 pig farms was 95.1%-99.9%,and the nucleotide similarity with reference strains such as JXA1,CH-1 a,NADC30,and FJFS was 92.1%-99.6%.The nucleotide similarity among PCV-2 ORF2 genes of 18 pig farms was 99.2%-99.9%,and the nucleotide similarity with reference strains such as BDH,AF055394,AF055392 and Zhuji2003 was 99.2%-99.6%.The nucleotide similarity among the PRV gE genes of 13 pig farms was 99.2%-99.8%,and the nucleotide similarity with reference strains such as HBCX2013,NYXY and Ea was 98.3%-99.7%.The res
