摘  要:为了研究兔出血症病毒2型(RHDV2)的基因组特性及遗传进化关系,试验对四川省某兔场死亡兔的组织病料进行RHDV2鉴定,采用RT-PCR方法扩增RHDV2全基因组分段序列,经DNAStar拼接获得病毒全基因组序列;采用MegAlign软件将分离株的全基因组、NSPs、VP60及VP10基因核苷酸序列和氨基酸序列与经典RHDV、RHDVa、RHDV2(RHDVb/GI.2)株进行了比对分析,并基于GenBank中44株RHDV的VP60基因构建分离株的系统发育树。结果表明:从病料组织中鉴定到RHDV2型毒株,命名为RHDV2-CHN/SC2020株,RHDV2-CHN/SC2020株与经典RHDV株、RHDVa株和RHDV2(RHDVb/GI.2)株的核苷酸序列相似性分别为84.6%~84.9%、83.4%~83.8%和92.0%~98.2%,与RHDV2型的相似性最高;RHDV2-CHN/SC2020株与荷兰株(RHDV2-NL2016)处于同一分支,亲缘关系最近。说明RHDV2已经传入我国,而遗传关系证实该病可能是从欧洲国家荷兰引种传入。In order to study the genomic characteristics and genetic evolution relationship of Rabbit hemorrhagic disease virus type 2(RHDV2)that occurred in China,the experiment was carried out to identify RHDV2 on infected tissue materials of dead rabbits in a rabbit farm in Sichuan Province.The RT-PCR method was used to amplify the complete gene sequence of RHDV2,and the complete genome sequence of the virus was obtained by DNAStar splicing;MegAlign software was used to analyze the homology between the complete genome,NSPs,VP60 gene,and VP10 gene nucleotide and amino acid sequences of the isolates and the classic RHDV,RHDVa,RHDV2(RHDVb/GI.2)strains;the phylogenetic tree of the isolates was constructed based on the VP60 genes of 44 RHDV strains in the GenBank database.The results showed that the RHDV2 strain was identified from the disease material tissue and named as RHDV2-CHN/SC2020 strain.The nucleotide homology between RHDV2-CHN/SC2020 and the classic RHDV,RHDVa and RHDV2(RHDVb/GI.2)strains were 84.6%-84.9%,83.4%-83.8%and 92.0%-98.2%,respectively,which meant the highest similarity with RHDV2.The RHDV2-CHN/SC2020 isolate and the Dutch strain(RHDV2-NL2016)existed in the same branch and were closely related.The results suggested that RHDV2 had been introduced into China,and genetic relationships had confirmed that the disease may be introduced from the Netherlands in Europe.
