摘 要:固醇调节元件结合蛋白1(Sterol regulatory element binding protein,SREBP1)作为一种核转录因子,与哺乳动物体内胆固醇及脂肪的生成密切相关。为了解牦牛乳腺组织中乳脂合成的机制及其特点,采集处于不同生理状态下(妊娠期、泌乳期、干奶期)的乳腺组织样品,采用qRT-PCR、Western-blot及免疫组化(immunohistochemistry,IHC)等技术检测SREBP1基因及其蛋白的相对表达情况。结果显示,泌乳期牦牛乳腺组织中SREBP1 m RNA表达量显著高于妊娠期和干奶期(P<0.05),且妊娠期SREBP1 m RNA表达量高于干奶期(P<0.05)。在牦牛不同生理状态下,SREBP1蛋白表达也发生了显著变化,妊娠期和泌乳期SREBP1蛋白表达量显著高于干奶期(P<0.05)。免疫组化法定位检测结果显示,SREBP1蛋白主要分布在牦牛乳腺腺泡上皮细胞、乳腺导管上皮细胞及血管内皮细胞。上述结果表明SREBP1及其编码m RNA的表达在牦牛乳腺组织中与其所处不同生理状态密切相关,妊娠期和泌乳期SREBP1的高表达,可能意味着其在牦牛乳腺发育和乳脂合成的过程中发挥着重要作用。本研究为进一步探明SREBP1在牦牛乳脂合成调控中的作用积累了一些基础资料。As a nuclear transcription factor,sterol regulatory element binding protein(SREBP1) is closely related to the generation of cholesterol and fat in mammals.To investigate the mechanism and characteristics of milk fat synthesis in yak breast tissue,the samples were collected at different physiological stages(pregnancy,lactation,dried milk period).qRT-PCR,Western-blot and immunohistochemistry(IHC) assays were used to study the relative expression of SREBP1 gene and its protein.The results showed that SREBP1 m RNA in breast tissue of yak during lactation was significantly higher than that of developmental and dried milk period(P<0.05),and the expression of SREBP1 m RNA during developmental was higher than dried milk period(P<0.05).The expression of SREBP1 protein also was changed significantly in breast tissue of yak during different physiological stages.The expression of SREBP1 protein during pregnancy and lactation period was significantly higher than dried milk period(P <0.05).Furthermore,the immunohistochemistry localization detection results showed that SREBP1 was mainly distributed in yak mammary gland acinar epithelial cells,mammary duct epithelial cells and vascular endothelial cells.These results indicated that the expression of SREBP1 and its coding m RNA in yak mammary tissue were closely related to its physiological stages,and the high expression of SREBP1 during pregnancy and lactation stage revealed that it played an important role in the development of yak mammary gland and milk fat synthesis.This study accumulated some basic data to further explore the role of SREBP1 in the regulation of yak milk fat synthesis.
