摘  要:为了探讨腱糖蛋白C(TNC)、磷脂酰肌醇-3,4,5-三磷酸依赖的Rac交换因子1(PREX1)、血小板凝血酶蛋白1(THBS1)和纤溶酶原激活物抑制因子1(SERPINE1)的m RNA在犬乳腺肿瘤中的表达水平及其对犬乳腺肿瘤的影响,采用荧光定量PCR技术对临床收集的24例犬乳腺肿瘤组织及其癌旁组织中TNC、PREX1、THBS1和SERPINE1的m RNA表达水平进行检测,并分析它们的表达水平与临床病例参数的关联性。荧光定量PCR结果显示:犬乳腺肿瘤组织与癌旁组织相比,TNC和THBS1的m RNA表达水平具有显著性差异(P<0.05),PREX1 m RNA表达水平具有极显著性差异(P<0.001),SERPINE1 m RNA的表达水平差异无统计学意义(P>0.05);TNC m RNA在犬乳腺组织中的表达水平与其他3种因子的m RNA表达水平无相关性(P>0.05);THBS1 m RNA与PREX1 m RNA、SERPINE1 m RNA的表达水平皆呈显著性正相关(P<0.05);PREX1 m RNA表达水平与SERPINE1 m RNA表达水平呈极显著性正相关(P=0.008);除了THBS1 m RNA在犬乳腺肿瘤中的表达水平与患犬品种之间具有显著性相关(P=0.011)外,这4种因子的m RNA在犬乳腺肿瘤中的表达水平与临床病例参数之间无显著性相关(P>0.05)。TNC、PREX1、THBS1和SERPINE1的m RNA在犬乳腺肿瘤中高表达,THBS1、PREX1和SERPINE1的m RNA在犬乳腺肿瘤中表达水平两两之间具有显著性相关,提示这3种因子的m RNA表达水平可能共同参与了犬乳腺肿瘤的进展,对诊断犬乳腺肿瘤有参考价值。To investigate the m RNA expression levels of tenascin-C(TNC),phosphatidylinositol-3,4,5-triphosphate-dependent Rac exchange factor 1(PREX1),thrombo-spondin 1(THBS1) and plasminogen activator inhibitor 1(SERPINE1) in canine mammary tumors and their effects on canine mammary tumors,the m RNA expression levels of TNC,PREX1,THBS1 and SERPINE1 in 24 canine mammary tumor tissues and their paracancerous tissues collected from clinical cases were examined by fluorescent quantitative PCR,and their correlation with clinical parameters was analysed.The results of fluorescent quantitative PCR showed that the m RNA expression levels of TNC and THBS1 were significantly different(P<0.05),PREX1 m RNA was highly significant(P<0.001) and SERPINE1 m RNA was not statistically significant(P>0.05) in canine mammary tumor tissues compared to paraneoplastic tissues.The expression levels of TNC m RNA in canine mammary tissue did not correlate with those of the other three factors(P>0.05);THBS 1 m RNA showed a significant positive correlation with PREX1 m RNA and SERPINE1 m RNA(P<0.05);PREX1 m RNA expression levels showed a highly significant correlation with SERPINE1.There was no significant correlation between the m RNA expression levels of these four factors in canine mammary tumors and clinical case parameters(P>0.05),except for THBS1 m RNA expression levels in canine mammary tumors,which were significantly correlated with the breed of affected dogs(P=0.011).The m RNA expression levels of TNC,PREX1,THBS1 and SERPINE1 were highly expressed in canine mammary tumors,and the m RNA expression levels of THBS1,PREX1 and SERPINE1 were significantly correlated between the two in canine mammary tumors,suggesting that the m RNA expression levels of these three factors may be jointly involved in the progression of canine mammary tumors and have reference value for the diagnosis of canine mammary tumors.
