摘 要:目的探究微小RNA-29b(miRNA-29b)对卵巢癌细胞生物学行为、抗生素核因子kB(NF-kB)和人核因子κB抑制蛋白α(IKB-α)的影响。方法将卵巢癌细胞株随机分为3组:第1转染组(卵巢癌细胞株不做其他处理,正常培养),第2转染组(将miR-29b空载体转染到卵巢癌细胞株中)和第3转染组(将miR-29b慢病毒转染到卵巢癌细胞中)。以荧光定量-PCR法检测卵巢癌细胞中miR-29b基因表达量,以蛋白质印迹法检测卵巢癌细胞中NF-KBp65和IKB-α的蛋白表达量。结果第1转染组、第2转染组和第3转染组的miR-29b基因表达量分别为0.45±0.08,0.46±0.09,0.89±0.34,组间比较差异有统计学意义(P<0.05),说明转染成功。第1转染组、第2转染组和第3转染组的NF-KBp65蛋白表达量分别为0.94±0.24,0.93±0.21,0.44±0.11;IKB-α蛋白表达量分别为0.38±0.09,0.37±0.10,1.02±0.41。第3转染组和第1转染组、第2转染组比较,上述指标的差异均有统计学意义(均P<0.05)。结论 miR-29b基因可能通过上调IKB-α表达水平进而抑制NF-KBp65活性,起到抑制卵巢癌细胞迁移的作用。Objective To explore the effects of microRNA-29 b(miRNA-29 b) on the biological behavior of ovarian cancer cells and the antibiotic nuclear factor κB(NF-κB) and inhibitor of human nuclear factor kappa B(IκB-α). Methods Ovarian cancer cell lines were randomly divide them into three groups: transfection-1 group(ovarian cancer cell lines were cultured normally without any other treatment), the transfection-2 group(the mir-29 b empty vector was transfected into ovarian cancer cell line), and the transfection-3 group(miR-29 b lentivirus was transfected into ovarian cancer cells). The expression of miR-29 b gene in ovarian cancer cells was detected by fluorescence quantitative-PCR method.The protein expression of NF-KBp65 and IKB-α in cancer cells was detected by Western blotting. Results The expression levels of miR-29 b gene in the transfection-1 group, transfection-2 group, and transfection-3 group were 0.45±0.08, 0.46±0.09, 0.89±0.34, respectively;the difference between the two groups was statistically significant(P<0.05),it indicating that the transfection was successful. The expression of the NF-KBp65 protein in the transfection-1 group,transfection-2 group,and transfection-3 group were 0. 94 ± 0. 24,0. 93 ± 0. 21,0. 44 ± 0. 11;and expression of the IKB-α protein were 0. 38 ± 0. 09,0. 37 ± 0. 10,1. 02 ± 0. 41. Comparison between transfection-3 group and transfection-1 group,transfection-2 group,the difference of factors were statistically significant( all P < 0. 05).Conclusion MiR-29 b may up regulate the expression of IKB-α,furthermore,NF-kBp65 activity was inhibited.It can inhibit the migration of ovarian cancer cells.
