摘 要:目的研究实验性糖尿病肾病SD大鼠模型建立的方法,并对其有效性进行评估。方法按照体重将SD大鼠随机分为2组:正常组和模型组,每组7只。给予大鼠一次性腹腔注射60 mg·kg^(-1)链脲佐菌素建立糖尿病肾病模型组。8周后,测定大鼠的随机血糖、肾指数、血酸肌酐水平、尿白蛋白水平和肾小球硬化指数。结果第8周,模型组和正常组随机血糖分别为(30.27±2.46),(5.76±1.06) mmol·L^(-1);肾指数分别为(0.66±0.06)%,(1.22±0.14)%;血酸肌酐水平分别为(0.64±0.08),(1.89±0.21) mg·dL^(-1);尿白蛋白水平分别为(3.24±0.61),(30.20±4.87) mg·dL^(-1);肾小球硬化指数分别为(6.23±2.10),(41.33±6.87)%。模型组与正常组比较,上述指标的差异均有统计学意义(均P<0.05)。结论本实验所建立的糖尿病肾病模型大鼠能引起肾功能紊乱及糖尿病肾病典型病理改变,表明成功复制实验性糖尿病肾病动物模型。Objective To study the method of establishing SD rat model of experimental diabetic nephropathy(DN) and evaluate its effectiveness. Methods SD rats were randomly divided into two groups according to weight: normal group and model group, with 7 rats in each group. The rats in model group were injected intraperitoneally with 60 mg·kg^(-1) streptozotocin to induce DN after fasting. After 8 weeks, the random blood glucose levels, renal index, serum creatinine level, urine albumin level, and glomerulosclerosis index were determined. Results The random blood glucose in model group and normal group were(30.27±2.46),(5.76±1.06) mmol·L^(-1);the renal index was(0.66±0.06),(1.22±0.14) %;the serum creatinine level was(0.64±0.08),(1.89±0.21) mg·dL^(-1);and the urinary albumin level was(3.24±0.61),(30.20±4.87) mg·dL^(-1);the glomerulosclerosis index was(6.23±2.10),(41.33±6.87)%. Comparison between model group and normal group, the difference of the factors were significant(all P<0.05). Conclusion The model of diabetic nephropathy was established can cause renal dysfunction and typical pathological changes of diabetic nephropathy and successfully establish experimental diabetic nephropathy animal model.
