摘  要:目的:建立HPLC多波长切换法同时测定苁蓉舒痉颗粒中松果菊苷、芍药苷、毛蕊花糖苷、丹酚酸B、丹皮酚和丹参酮ⅡA的含量。方法:采用Agilent Zorbax Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸溶液为流动相,梯度洗脱,流速1.0 mL·min^(-1),柱温25℃,进样量20μL,检测波长分别为330 nm(松果菊苷和毛蕊花糖苷)、230 nm(芍药苷)、286 nm(丹酚酸B和丹皮酚)、270 nm(丹参酮ⅡA)。结果:松果菊苷、芍药苷、毛蕊花糖苷、丹酚酸B、丹皮酚和丹参酮ⅡA质量浓度分别在18.18~181.8μg·mL-1(r=0.9998)、28.86~288.6μg·mL^(-1)(r=0.9998)、0.5760~5.760μg·mL-1(r=0.9995)、32.96~329.6μg·mL-1(r=0.9997)、31.97~319.7μg·mL^(-1)(r=0.9990)、1.120~11.20μg·mL^(-1)(r=0.9998)范围内与各峰面积呈良好的线性关系;平均回收率(n=6)分别为98.7%、99.4%、98.6%、99.0%、98.8%、99.1%,RSD分别为1.0%、0.92%、1.6%、0.85%、1.0%、1.7%;3批样品中上述各成分含量范围分别为3.571~3.702、6.692~6.911、0.071~0.089、7.343~7.947、2.201~2.259、0.041~0.055 mg·g^(-1)。结论:该方法可作为苁蓉舒痉颗粒质量标准同时测定6个主要成分的含量测定法。Objective:To establish an HPLC multi-wavelength switching method for the simultaneous determination of echinacoside,paeonia lactiflora,verbascoside,salvianolic acid B,paeonol,and tanshinoneⅡA in Congrong Shujing granules.Methods:An Agilent Zorbax Eclipse XDB-C18 column(250 mm×4.6 mm,5μm)was used,with acetonitrile-0.1%phosphoric acid solution as the mobile phase in gradient elution.The flow rate was 1.0 mL·min^(-1),the column temperature was 25℃,the injection volume was 20μL,and the detection wavelengths were set at 330 nm(echinacoside and verbascoside),230 nm(paeoniflorin),286 nm(salvianolic acid B and paeonol)and 270 nm(tanshinoneⅡA).Results:Echinacoside,paeoniflorin,verbascoside,salvianolic acid B,paeonol and tanshinoneⅡA showed good linearity in the ranges of 18.18-181.8μg·mL-1(r=0.9998),28.86-288.6μg·mL^(-1)(r=0.9998),0.5760-5.760μg·mL^(-1)(r=0.9995),32.96-329.6μg·mL^(-1)(r=0.9997),31.97-319.7μg·mL^(-1)(r=0.9990),1.120-11.20μg·mL^(-1)(r=0.9998),respectively.The average recovery(n=6)were 98.7%,99.4%,98.6%,99.0%,98.8%,99.1%with RSDs of 1.0%,0.92%,1.6%,0.85%,1.0%,and 1.7%,respectively.The content ranges of the above-mentioned components in 3 samples were 3.571-3.702 mg·g-1,6.692-6.911 mg·g^(-1),0.071-0.089 mg·g^(-1),7.343-7.947 mg·g^(-1),2.201-2.259 mg·g^(-1)and 0.041-0.055 mg·g^(-1),respectively.Conclusion:This method can be used as the quality standard of Congrong Shujing granules to simultaneously determine the contents of six main components.
