摘  要:目的:建立川芎药材HPLC指纹图谱及有效成分含量测定方法,用于川芎药材质量控制标准的提高。方法:对18批川芎药材进行HPLC指纹图谱构建。基于多元统计分析(主成分分析、聚类分析)综合评价18批药材的质量差异,探究影响质量差异的主要色谱峰,并对其有效成分进行含量测定。色谱条件:采用Kromasil C18色谱柱(250 mm×4.6 mm,5μm),以乙腈为流动相A,以0.1%磷酸水溶液为流动相B,进行梯度洗脱,流速1.0 mL·min^(-1),柱温30℃,检测波长为280 nm。结果:指纹图谱共标定了13个共有峰,通过比对指认出7个色谱峰。18批药材相似度均大于0.97。主成分得分结果与聚类分析结果一致,都将样品分为3类。18批川芎药材中阿魏酸、洋川芎内酯I、洋川芎内酯A、藁本内酯含量范围分别为0.17%~0.41%、0.08%~0.21%、0.63%~3.82%、1.02%~2.12%。结论:建立的分析方法稳定可靠,较2020年版《中华人民共和国药典》川芎药材质量标准更具有可控性。Objective:To establish HPLC fingerprint and determination method of active ingredients in Chuanxiong Rhizoma,which can be used to improve the quality control standard of Chuanxiong Rhizoma.Methods:The fingerprints of 18 batches of Chuanxiong Rhizoma were constructed.Based on multivariate statistical analysis(principal component analysis and cluster analysis),the quality differences of 18 batches of medicinal materials were comprehensively evaluated,the main chromatographic peaks affecting the quality differences were explored,and the content of their active ingredients was determined.Kromasil C18(250 mm×4.6 mm,5μm)was used as chromatographic column with acetonitrile as mobile phase A and 0.1%phosphoric acid solution as mobile phase B for gradient elution at a flow rate of 1 mL·min^(-1).The column temperature was 30℃and the detection wavelength was 280 nm.Results:13 common peaks were identified by fingerprint,and 7 peaks were identified by comparison.The similarities of 18 batches of herbs were above 0.97.The results of principal component scores were consistent with those of cluster analysis,and the samples were divided into three categories.The contents of ferulic acid,senkyunolide I,senkyunolide A and ligustilide in 18 batches of Chuanxiong Rhizoma were 0.17%-0.41%,0.08%-0.21%,0.63%-3.82%and 1.02%-2.12%,respectively.Conclusion:The established analytical method is stable and reliable,which is more controllable than the quality standard of Chuanxiong Rhizoma in Chinese Pharmacopoeia(2020 Edition).
