摘 要:目的通过网络药理学探讨知母皂苷A-Ⅲ抗鼻咽癌(NPC)的作用靶点及分子机制。方法利用Swiss Target Prediction数据库获取知母皂苷A-Ⅲ作用靶点,通过GeneCards数据库检索NPC相关疾病靶点,将其与知母皂苷A-Ⅲ作用靶点映射,使用Cytoscape 3.6.1软件构建知母皂苷A-Ⅲ-鼻咽癌疾病靶点网络图,并运用DAVID数据库进行GO功能富集分析及KEGG通路富集分析。结果获得知母皂苷A-Ⅲ抗鼻咽癌的潜在靶点31个,化合物-关键靶点-通路网络图显示关键靶点主要包括PRKCA、IGF1R、VEGFA和KDR等。GO功能富集分析获得分子功能条目13个、生物过程条目55个和细胞组成条目10个,KEGG通路富集得到20条相关通路,涉及蛋白多糖通路、癌症信号通路和PI3K-Akt信号通路等信号通路。结论基于网络药理学多靶点、多通路的整体作用特点,预测了知母皂苷A-Ⅲ抗鼻咽癌的作用靶点和潜在作用机制,以期为下一步实验研究提供科学依据。Objective To explore the acting targets and molecular mechanism of timosaponin A-Ⅲagainst nasopharyngeal carcinoma(NPC)through network pharmacology.Methods The Swiss Target Prediction database was utilized to obtain the acting targets of timosaponin A-Ⅲ,and the GeneCards database was searched for NPCrelated disease targets,which were mapped to the acting targets of timosaponin A-Ⅲ.Cytoscape 3.6.1 software was used to construct the disease target network diagram of timosaponin A-Ⅲ-NPC,and DAVID database was adopted to carry out GO functional enrichment analysis and KEGG pathway enrichment analysis.Results 31 potential targets of timosaponin A-Ⅲagainst NPC were obtained.The compound-key targets-pathway network diagram showed that the key targets mainly included PRKCA,IGF1R,VEGFA,KDR and so on.13 molecular function entries,55 biological process entries and 10 cell component entries were obtained by GO functional enrichment analysis.KEGG pathway enrichment yielded 20 related pathways,involving proteoglycan pathway,cancer signaling pathway,PI3K-Akt signaling pathway and other signaling pathways.Conclusion Based on the overall acting characteristics of multitargets and multi-pathways in network pharmacology,the acting targets and potential acting mechanism of timosaponin A-Ⅲagainst NPC have been predicted,in order to provide a scient ific basis for further experimental study.
