摘  要:试验以干旱盐碱地优势物种长穗柽柳的叶片、嫩茎、嫩根为外植体材料,在筛选最适宜其愈伤组织诱导材料的基础上,探讨不同培养基、外源激素种类(6-BA、2,4-D、IBA、NAA)和大量元素CaCl2浓度变化对不定芽诱导、增殖及生根的影响,建立其组织培养植株再生体系。结果表明:1)嫩茎是最适宜长穗柽柳进行愈伤组织诱导的外植体,在MS+1.5mg·L^(-1)6-BA+0.3mg·L^(-1)IBA的培养基配方中,出现诱导率最大值65.33%。2)MS+1.5mg·L^(-1)6-BA+0.05mg·L^(-1)NAA是最适宜进行不定芽诱导的培养基配方,诱导率最大值为57.32%。3)在MS+3.0mg·L^(-1)6-BA+0.5mg·L^(-1)2,4-D的培养基中不定芽增殖率为最大值63.29%,增殖数量多且生长健壮。4)最适宜的生根培养基为1/2 MS+1/2 CaCl2+3.0mg·L^(-1)IBA,最早13 d开始生根,平均根长3.52 cm,生根率82.16%,瓶苗主根长、毛根较多、长势好,有利于后期炼苗和移栽定植。The effects of different media,exogenous hormone types(6-BA,2,4-D,IBA,NAA)and the concentrations of CaCl2 on adventitious bud induction,proliferation and rooting were studied,and the plant regeneration system of tissue culture was established on the basis of selecting the most suitable materials for callus induction,and the leaves,tender stems and roots of Tamarix elongate,which is a dominant species in arid saline alkali soil,were used as explants.The results showed that:1)Tender stem is the most suitable explant for callus induction of T.elongata,and the highest induction rate was 67.15%in MS+1.5 mg·L^(-1)6-BA+0.3 mg·L^(-1)IBA.2)Medium formulation of MS+1.5 mg·L^(-1)6-BA+0.05 mg·L^(-1)NAA was the most suitable for adventitious bud induction,and the maximum induction rate was 57.32%.3)The adventitious bud proliferated in large quantity and grew well,and their proliferation rate reached the maximum of 63.29%in the medium of MS+3.0 mg·L^(-1)6-BA+0.5 mg·L^(-1)2,4-D.4)The most suitable rooting medium was 1/2 MS+1/2 CaCl2+3.0 mg·L^(-1)IBA.Root initiated in the earliest 13 days.The rooting rate was 82.16%and the average root length was 3.52 cm.The bottle seedlings had long main root,more hairy roots,and good growth vigor were conducive to the later stage of seedling refining,transplanting and planting.
