摘 要:[目的]探究miRNA-143-3P在从江香猪与杂交猪之间是否存在表达水平的差异性,预测分析该miRNA的靶基因及其功能,分析miR-143-3P在这两个猪种中的功能。[方法]通过RT-qPCR检测从江香猪与杂交猪中不同组织的miRNA-143-3P,利用线软件预测miRNA-143-3P的靶基因,对筛选的靶基因作生物信息学分析,了解miRNA-143靶基因的功能。[结果]荧光定量PCR检测的结果显示,在从江香猪中的表达量最高的是肝脏,而杂交猪的是脾脏,存在表达差异极显著的组织有肺、大肠、小肠、脾、肝(P <0.01)。软件预测靶基因时,共存的有30个。生物功能分析时发现,这些基因多数存在于细胞质(53.6%)和细胞核(46.4%)中,而在蛋白质结构域是在跨膜区和螺旋区占多数,分别占26.9%、23.1%,而在与转录因子结合较多的是SP1(71.4%),组织表达较高的为肺、肝、肾,高达80%,而热图显示RAB7A、LASP1、MYO6、KRAS这四个基因在数据库中的富集相对较广。[结论]miRNA-143-3P在从江香猪与杂交猪之间存在表达性差异,这种差异与该miRNA-143-3P所对应的靶基因功能相关。[Objective]To explore whether miRNA-143-3P expression levels are different between Conjiang xiang pigs and hybrid pigs,and analyze the functions of target genes of this miRNA in advance,and analyze the functions of miR-143-3P in these two pig species.[Method]miRNA-143-3P was detected in different tissues from Jiangxiang pigs and hybrid pigs by RT-qPCR,and the target genes of miRNA-143-3P were predicted by line software,and the screened target genes were analyzed by bioinformatics to understand the functions of miRNA-143 target genes.[Result]The fluorescence quantitative PCR results showed that liver was the most highly expressed pig in concomitant pigs,while spleen was the most expressed in hybrid pigs. The tissues with significant differences in expression included lung,large intestine,small intestine,spleen and liver(P <0. 01). When the software predicted the target genes,30 coexisted. Biological function analysis found that the most genes exist cytoplasm(53. 6%) and the nucleus(46. 4%),and in the protein structure domain is across the membrane area and screw area accounted for 26. 9%,23. 1% respectively,and a majority in combine with transcription factors is more SP1(71. 4%),organizations expressed higher as the lung,liver,kidney,as high as 80%,and heat maps show RAB7 A,LASP1,MYO6,KRAS enrichment of the four genes in the database is relatively wide.[Conclusion]There was a difference in the expression of miRNA-143-3P between conjiang pigs and hybrid pigs,and this difference was related to the function of the target gene corresponding to the miRNA-143-3P.
