摘 要:[目的]分析miR-137靶向调控EZH2的作用,及卵巢癌中血清miR-137对疾病预后的预测价值。[方法]将本院2018年1月~2019年6月收治的100例卵巢癌患者作为研究对象,使用SW626细胞系分为miR-NC组、miR-137组和miR-137+EZH2组;采用MTT法检测细胞增殖活性,Western Blot法检测细胞中EZH2蛋白水平,靶基因双荧光素酶报告基因检测分析miR-137与EZH2的调控关系。荧光实时定量qRT-PCR法检测细胞中EZH2、GAPDH水平。所有患者入组后采集空腹静脉血,采用qRT-PCR法检测受试者血中miR-137水平。[结果]miR-NC组、miR-137组及miR-137+EZH2组细胞增殖活性存明显差异,其中miR-137组细胞增殖活性最低,且差异存统计学意义(P <0.05);miR-NC组、miR-137组及miR-137+EZH2组细胞中EZH2蛋白和mRNA表达水平存明显差异,其中EZH2蛋白和mRNA表达水平最低,且差异存统计学意义(P <0.05);卵巢癌SW626细胞转染mut-EZH2 3’-UTR、wt-EZH2 3’-UTR载体后,其野生型双荧光素酶活性分别为1.023±0.205和2.563±0.314,均显著低于突变型3.848±0.632和4.104±0.647,且差异存统计学意义(P <0.05);存活组患者血中miR-137水平明显高于死亡组,且差异存统计学意义(P <0.05);miR-137鉴别评估卵巢癌患者预后质量的敏感度、特异度均> 85%,AUC高于0.90。[结论]miR-137可靶向调控卵巢癌细胞中EZH2表达,且卵巢癌患者血清miR-137对疾病预后质量预测具有较高的应用价值。[Objective]To analyze the role of miR-137 in targeted regulation of EZH2,and the predictive value of serum miR-137 in ovarian cancer for the prognosis of the disease.[Method]100 ovarian cancer patients from January 2018 to June 2019 in our hospital were selected as the research objects;SW626 cell line was used to divide into miR-NC group,miR-137 group and miR-137 + EZH2 group;MTT method was used to detect cell proliferation activity,Western Blot method was used to detect the level of EZH2 protein in cells,and the target gene dual luciferase reporter gene detection was used to analyze the regulatory relationship between miR-137 and EZH2. Fluorescence real-time quantitative qRT-PCR method was used to detect the levels of EZH2 and GAPDH in cells. Fasting venous blood was collected from all patients after enrollment,and the level of miR-137 in the subjects’ blood was detected by qRT-PCR.[Result]There were significant differences in cell proliferation activity among mir-nc group,miR-137 group and miR-137 + EZH2 group,among which miR-137 group had the lowest cell proliferation activity(P < 0. 05);The expression levels of EZH2 protein and mRNA in mir-nc group,miR-137 group and miR-137 + EZH2 group were significantly different,and the expression levels of EZH2 protein and mRNA were the lowest(P <0. 05);After transfection of mut-ezh2 3’-UTR and wt-ezh2 3’-UTR vectors into sw626 cells,the wild-type dual luciferase activities were 1. 023 ± 0. 205 and 2. 563 ± 0. 314,respectively,which were significantly lower than those of mutant 3. 848 ± 0. 632 and 4. 104 ± 0. 647(P < 0. 05);The level of miR-137 in the survival group was significantly higher than that in the death group(P < 0. 05);The sensitivity and specificity of miR-137 in differential evaluation of prognosis quality of ovarian cancer patients were more than 85%,and the AUC was higher than 0. 90.[Conclusion]miR-137 can target and regulate the expression of EZH2 in ovarian cancer cells,and serum miR-137 in patients with ovarian cancer has high application value
