摘 要:目的:研究缩泉益肾方与益智仁对糖尿病肾脏疾病(DKD)小鼠疗效及肠道菌群的影响差异。方法:以自发型糖尿病肾脏疾病(db/db)小鼠为动物模型,随机分为益智仁组、缩泉益肾方组及生理盐水组,每组8只。3组小鼠分别给予1.5 mg/g益智仁、0.01 mg/kg缩泉益肾方及生理盐水,连续灌胃8周。测定小鼠血糖、血脂及氧化应激指标;收集小鼠粪便,测定粪便菌群的多样性、丰富度及表达差异,比较3组厚壁菌门与拟杆菌门的比值、厚壁菌门、Akkermansia菌及拟杆菌的丰富度。结果:与生理盐水组血糖、甘油三脂、CAT、H_(2)O_(2)及O^(2-)相比,缩泉益肾方组与益智仁组均明显降低,差异具有统计学意义(P<0.05或P<0.01),且缩泉益肾方效果优于益智仁;与生理盐水组相比,缩泉益肾方与益智仁均能调节厚壁菌门与拟杆菌丰度比、厚壁菌门、拟杆菌及Akkermansia丰富度。结论:推测益智仁与缩泉益肾方调节DKD的作用可能与调节血糖、血脂、氧化应激反应及肠道微生物有关,且缩泉益肾方作用较益智仁作用强。Objective:To compare the effects of Sou-quan-yi-shen prescription and Yizhiren on the efficacy and gut microbiome of DKD mice.Methods:Taking spontaneous diabetic kidney disease(db/db)mice as the animal model,the mice were randomly divided into Yizhiren group,Sou-quan-yi-shen prescription group,saline group,each with 8 mice.Three groups of mice were given 1.5 mg·g-1 Yizhiren,0.01 mg·kg-1 Sou-quan-yi-shen prescription and saline,continuous gavage for 8 weeks.Blood glucose,blood lipids and oxidative stress were measured.The fecal gut microbiome,richness and expression difference were determined by collecting mouse feces,and the ratio of Firmicutes to Bacteroidetes,as well as the richness of Firmicutes,Akkermansia and Bacteroidetes in the three groups of mice were compared.Results:Compared with the normal saline group of blood glucose,TG,CAT,H_(2)O_(2) and O^(2-),the Sou-quan-yi-shen prescription group and the Yizhiren group were significantly lower(P<0.05 or P<0.05),and the effect of Sou-quan-yi-shen prescription is better than Yizhiren.Compared with the normal saline group,Souquan-yi-shen prescription and Yizhiren can adjust the abundance ratio of Firmicutes and Bacteroides,Firmicutes,Bacteroides and Akkermansia.Conclusion:The effect of Sou-quan-yi-shen prescription and Yizhiren on regulating DKD may be related to the regulation of blood sugar,blood lipids,oxidative stress and gut microbiome,and the effect of Sou-quan-yi-shen prescription is better than that of Yizhiren.
