摘 要:目的明确核磁共振(Magnetic Resonance,MR)对比剂钆喷替酸葡甲胺对软骨细胞凋亡、自噬的影响。方法体外培养人软骨细胞,分为正常对照组、低剂量组、中剂量组及高剂量组,CCK8检测各组细胞增殖情况;流式细胞仪检测各组细胞凋亡率;Western blot检测各组细胞凋亡相关蛋白bcl2、bax、cleaved-caspase3及自噬相关蛋白lc3、p62蛋白等表达。结果低剂量组、中剂量组与正常对照组细胞增殖率比较无统计学差异(P>0.05),高剂量组与正常对照组比较抑制细胞增殖(P<0.05);低剂量组、中剂量组与正常对照组细胞凋亡率比较无统计学差异(P>0.05),高剂量组与正常对照组比较细胞凋亡率增高(P<0.05);低剂量组、中剂量组与正常对照组比较,bcl2、bax、cleaved-caspase3蛋白表达差异无统计学意义(P>0.05),高剂量组与正常对照组比较,bcl2蛋白表达下调,bax、cleaved-caspase3蛋白表达上调,差异均具有统计学意义(P<0.05)。低剂量组、中剂量组与正常对照组比较,lc3Ⅱ蛋白表达上调,p62蛋白表达下调(P<0.05),高剂量组与正常对照组比较,lc3Ⅱ蛋白表达下调,lc3、p62蛋白表达上调,差异均具有统计学意义(P<0.05)。结论低浓度钆喷替酸葡甲胺促进人软骨细胞自噬,而高浓度钆喷替酸葡甲胺能够抑制人软骨细胞增殖及自噬,促进软骨细胞凋亡。ObjectiveTo compare the effect of Magnetic Resonance(MR)and contrast agent gadopentine meglumine on the proliferation and apoptosis of chondrocytes.MethodsHuman chondrocytes were cultured in vitro and they were divided into control group,low-dose group,middle-dose group and high-dose group.Cell proliferation in each group was detected by CCK8.Cell apoptosis rate in each group was detected by flow cytometry.The expression of apoptosis-related proteins bcl2,bax,cleaved-caspase3 and autophagy-related proteins lc3,p62 proteins were detected in each group.ResultsThere was no significant differences between the low-dose group,the middle-dose group and the normal control group on cell proliferation rate(P>0.05).Compared with normal control group,the cell proliferation rate was significantly depressed in high-dose group(P<0.05).There was no significant difference between the low-dose group,the middle-dose group and the normal control group on cell apoptosis rate(P>0.05).Compared with normal control group,the cell apoptosis rate was significantly increased in high-dose group(P<0.05).The expression of bcl2,bax and cleaved-caspase3 were not significantly difference between the low-dose group,the middle-dose group and the normal control group(P>0.05).The expression of bcl2 protein was down-regulated,and the expression of bax and cleaved-caspase3 protein was up-regulated in high-dose group(P<0.05).Compared with the normal control group,the expression of lc3Ⅱprotein was up-regulated,and the expression of p62 protein was down-regulated significantly in the low-dose group and the middle-dose group(P<0.05).Compared with the normal control group,the expression of lc3Ⅱprotein was down-regulated and the expression of lc3 and p62 protein was up-regulated in high-dose group(P<0.05).ConclusionLow concentration of gadopentine meglumine could promote autophagy of human chondrocytes,while high concentration of gadopentine meglumine can inhibit human chondrocyte proliferation and autophagy,and promote chondrocyte apoptosis.
