摘  要:研究了从红缘拟层孔菌中分离纯化的3-乙酰氧基-8,24-羊毛甾二烯-21酸(FPOA)对人乳腺癌细胞MCF-7细胞增殖的抑制作用及诱导凋亡的作用机理。采用MTT法检测细胞抑制增殖的能力,通过Annexin V-FITC/PI双染法检测细胞凋亡情况,采用蛋白质印迹法分析凋亡相关蛋白的表达情况。结果表明,MCF-7经FPOA处理后培养12、24、48 h,细胞毒活性明显,IC50值分别为62.36,27.06和23.36#g/mL。经质量浓度为50μg/mL的FPOA处理24 h后,细胞凋亡率达到39.58%。蛋白检测发现,Cytochrome c和Caspase-3的含量明显升高,Bcl-2/Bax的表达明显降低。说明FPOA能够调节凋亡相关蛋白(Cytochrome c,Caspase-3,Bcl-2和Bax)的表达量,从而介导凋亡;推测FPOA诱导MCF-7细胞凋亡的作用机制与线粒体途径相关。The mechanism of cytotoxicity and apoptosis of 3-acetoxylanosta-8,24-dien-21-oic acid( FPOA) on MCF-7 cells was studied. The cytotoxicity to MCF-7 was detected by MTT assay. Apoptosis ability was tested through AnnexinV-FITC/PI double staining. The expression of apoptosisrelated proteins was analyzed by western-blotting. The results showed that cytotoxicity of FPOA on MCF-7 cells was obvious. The IC50 values were 62.36,27.06 and 23.36 μg/mL after 12,24 and 48 hours treated by FPOA,respectively. The apoptotic rate reached 39.58% with 50 μg/mL FPOA after 24 hours treatment. The expression of Cytochrome C and Caspase-3 increased significantly,while the ratio of Bcl-2/Bax decreased evidently in protein detection. The expression of apoptosisrelated proteins was regulated to induce apoptosis,such as Cytochrome c,Caspase-3,Bcl-2 and Bax. Therefore,it demonstrated that the mitochondria pathway was the apoptosis mechanism of MCF-7 cells induced by FPOA.
