摘 要:目的:探讨芹黄素对脑缺血再灌注损伤沙土鼠的脑保护作用及其与基质金属蛋白酶9的相关性。方法:将40只清洁级健康沙土鼠随机分为假手术组、模型组、尼莫地平组(2 mg/kg)、芹黄素组(40 mg/kg),每组各10只。采用双侧颈总动脉夹闭手术法建立脑缺血再灌注损伤沙土鼠模型;于建模前1周开始腹腔注射给药,在建模手术后6 h内记录各组神经症状并做卒中评分,6 h后免疫组化法测定脑组织海马CA1区基质金属蛋白酶9的表达。结果:与假手术组比较,模型组卒中评分显著提高(P<0.01);与模型组比较,芹黄素组与尼莫地平组评分均显著降低(P<0.01);与假手术组比较,模型组沙土鼠海马CA1区基质金属蛋白酶9的表达IOD值明显升高(P<0.01);与模型组比较,芹黄素组、尼莫地平组海马CA1区基质金属蛋白酶9表达均明显降低,组间比较差异有统计学意义(P<0.01)。结论:芹黄素对脑缺血再灌注损伤沙土鼠具有脑保护作用,可能与其能抑制基质金属蛋白酶9的活化有关。Objective:To approach the protective effect of apigenin on brain of gerbils with cerebral ischemia/reperfusion injury and its preliminary mechanism.Methods:40 healthy gerbils of clean grade were randomly divided into sham operation group,model group,nimodipine group(2 mg/kg),apigenin group(40 mg/kg),10 gerbils in each group.The model of gerbil with cerebral ischemia/reperfusion injury was prepared by a surgery of clipping bilateral common carotid artery.The administration by intraperitoneal injection was started at 1 week before modeling.Within 6 hours after modeling operation,the neurological symptoms of each group were recorded,and the stroke was scored.After 6 hours,the expression of matrix metalloproteinase-9 in hippocampal CA1 area of brain tissue was detected by immunohistochemistry.Results:Compare with sham operation group,the stroke score of model group was significantly enhanced(P﹤0.01).Compared with model group,the stroke scores of apigenin group and nimodipine group were significantly reduced(P﹤0.01).Compare with sham operation group,IOD value of matrix metalloproteinase-9 expression of hippocampal CA1 area in gerbils of model group was significantly enhanced(P﹤0.01).Compared with model group,the expressions of matrix metalloproteinase-9 of hippocampal CA1 area in apigenin group and nimodipine group were significantly reduced,and the difference compared between two groups was statistically significant(P﹤0.01).Conclusion:Apigenin has a protective effect on brain of gerbils with cerebral ischemia/reperfusion injury,which may be related to apigenin inhibiting the activation of matrix metalloproteinase-9.
