“如源一号”治疗猴免疫缺陷病毒感染恒河猴的药效观察

摘  要:目的评价"如源一号"治疗猴免疫缺陷病毒(SIV)感染中国恒河猴的效果。方法选取经静脉注射感染SIVmac239的中国恒河猴15只,随机分为3组,分别为对照组和"如源一号"低剂量组、高剂量组。检测不同时期血常规、CD4细胞和CD8细胞亚群、SIV病毒载量的变化,并活检取淋巴结观察病理学的情况。结果 SIV感染后对照组的体重持续下降;而高、低剂量组的体重略有增加,给药8周时高剂量组的平均体重增加值为0.28 kg,优于对照组的-0.22 kg(P <0.05)。各组病毒载量、CD4细胞及CD4/CD8细胞比值维持稳定;低剂量组在停药8周期间,病毒载量平均下降了0.63 log_(10) copies/mL(P>0.05)。对照组的淋巴结组织结构持续破坏、退化;而高剂量组动物的淋巴结结构一直维持于增生的免疫增强状态;高、低剂量组在停药后淋巴结的结构均继续维持稳定、甚至低剂量组持续改善。结论 "如源一号"能改善SIV感染猴体重,且对病毒载量控制、淋巴结构的改善具有一定的保护作用。Objective To evaluate the therapeutic efficacy of Ru Yuan Yi Hao(Restore Plus Granules, RPG) on simian immunodeficiency virus(SIV) infected monkeys. Methods Fifteen Chinese rhesus monkeys were infected with SIVmac239 intravenously and randomly assigned into three groups, namely the control group, the low-dose RPG treatment group and the high-dose RPG treatment group, with 5 monkeys in each group. In pre-designed time-points, the blood routine analysis was performed, and their CD4^(+) and CD8^(+)T lymphocyte subsets proportion and counts, as well as their plasma SIV loads, were determined. Moreover, the pathological examination and scoring were performed with biopsy of their lymph nodes. Results The body weight in the control group declined persistently, while that in high-and low dose treatment groups increased slightly. At 8 weeks of administration, the weight gain of the high-dose group was 0.28,better than-0.22 of the control group(P<0.05). The change of body weight of the high-dose treatment group was significantly improved compared with that of the model group at 8 weeks after RPG administration(P<0.05). Compared with that of the control group, the viral load of the treatment groups was slightly inhibited after treatment, and the CD4^(+)/CD8^(+)T cell ratio remained stable. Viral load in the low-dose group decreased by an average of 0.63 log_(10) copies/mL during8 weeks of withdrawal(P > 0.05). Besides, after treatment, the lymph node structure of 4 monkeys in the low-dose group was "improved", and that of 3 monkeys in the high-dose group remained in the "proliferative" state, and the improvement persisted even after the withdrawal of treatment. Conclusion RPG can improve the body weight of SIV infected monkeys, suppress the virus replication slightly, and protect the lymph node from damage to some extent.
