摘  要:目的:运用Box-Behnken响应面法优化滇黄精产地加工炮制一体化工艺。方法:采用蒽酮-硫酸紫外分光光度法检测,黄精多糖含量为评价指标。单因素试验确定因素最低水平和最高水平,结合Box-Behnken中心组合设计,对滇黄精蒸制时间、切片厚度、干燥温度3个因素进行优化。结果:滇黄精产地加工炮制一体化优化工艺为蒸制时间22 min、切片厚度3.6 mm、干燥温度61℃。优化工艺检测黄精多糖质量分数为12.12%,与预测值相接近,实验所建模型准确可靠。结论:Box-Behnken响应面优化滇黄精产地加工炮制一体化工艺切实可行,为筛选适宜滇黄精产地加工炮制一体化方法研究提供参考。Objective:To optimize integrative primary processing technology of Polygonatum kingianum by Box-Behnken response surface methodology.Methods:The anthrone-sulfuric acid method was used to determine the polysaccharide content of P.kingianum.The single factor test determined the lowest and highest level of factors first,then we optimized the steaming time,slice thickness and drying temperature of P.sibiricum by the Box-Behnken combination design.Results:Optimum parameters for primary processing were as follow:steaming time of 22 min,slice thickness of 3.6 mm,and drying temperature of 61℃.The actual content of P.sibiricum polysaccharide was 12.12%,which was close to the predicted value.Conclusion:It is suitable to use the Box-Behnken response surface method to optimize the integrative primary processing of P.kingianum,the study provides a basis for the research on the integrative technology of primary processing of P.kingianum.
