朱砂及其复方与氯化汞、甲基汞对小鼠肾转运体基因表达影响对比

摘　　要：目的:对比研究朱砂及其复方与其他含汞化合物对小鼠肾脏转运体基因表达的影响。方法:清洁级健康雄性成年昆明种小鼠分为生理盐水、朱砂安神丸1.8 g·kg^(-1)(含汞0.17 g·kg^(-1))、朱砂0.2 g·kg^(-1)(含汞0.17 g·kg^(-1))、高剂量朱砂2g·kg^(-1)(含汞1.7 g·kg^(-1))、HgS 0.2 g·kg^(-1)(含汞0.17 g·kg^(-1))、HgCl_(2)0.032 g·kg^(-1)(含汞0.024 g·kg^(-1),约1/7朱砂中汞含量)、MeHg 0.026 g·kg^(-1)(含汞0.024 g·kg^(-1),约1/7朱砂中汞含量)组,每天给药1次,连续30 d。30 d后处死各组小鼠取肾脏,RT-PCR方法检测小鼠肾组织有机阴离子转运体多肽(Oatp4c1)、胆酸转运体(Ost-α、Ost-β)的表达。结果:与正常组相比,HgCl_(2)组和MeHg组Ost-α、Ost-β基因的表达显著降低(P<0.05);朱砂安神丸组Oatp4c1基因的表达显著升高(P<0.05)。结论:HgCl_(2)组和MeHg组小鼠肾转运体基因表达与正常组相比有显著性差异,朱砂及朱砂安神丸对肾转运体基因表达影响较小。Objective Compares the effects of cinnabar and its compound with other mercury-containing compounds on the expression of renal transporter genes in mice.Methods Healthy male mice were gavaged with equivalent physiological saline,Zhusha Anshenwan 1.8 g·kg^(-1),cinnabar 0.2 g·kg^(-1),high dose cinnabar 2 g·kg^(-1),HgS 0.2 g·kg^(-1),HgCl_(2)0.032 g·kg^(-1),MeHg 0.026 g·kg^(-1),mnce daily,for 30 d,and kidney toxicity was detertmined by measuring body mass gain.Kill the mice and take the kidney after 30 days.The mRNA levels of Oatp4c1,Ost-αand Ost-βwere detected with RT-PCR.Results Compared with normal control group,mRNA expressions of Ost-αand Ost-βwere evidently lower in HgCl_(2)and MeHg groups(P<0.05).However mRNA expressions of Oatp4c1 were apparently higher in Zhusha Anshenwan group than in control group(P<0.05).Conclusion The expression of mRNA of Ost-αand Ost-βhave a significant changes in HgCl_(2)and MeHg groups.But most of them were not changed in other groups.The mRNA change in some kidney transporters may be related to the Pharmacological effects.
